Inter-simple sequence repeat (ISSR) markers were used to investigate the genetic diversity of 13 wild Auricularia polytricha strains collected from Guangdong Province and six cultivars from Henan, Fujian, Guangdong and Hainan. Eight primers produced a total of 265 loci in 19 strains, and the percentage of polymorphic loci was 99%. The mean genetic similarity of wild A. polytricha was 75%, and 72. 3% in cultivated strains, suggested the genetic variation of cultivars was slightly higher than that intraregional wild strains. Clustering analysis based on genetic distance revealed the 19 strains could be classified into three distinct groups. Group I composed all the wild strains, Group II included three cultivars from Henan and Guangdong that had a closer relationship with the wild strains, and the group HI comprised the other three cultivars. The results of principal coordinate analysis were in accordance with the neighbor-joining clustering, and the first three most informative coordinate accounted for 74.3% of the variations, indicated a high level of genetic diversity among A. polytricha strains.
