Identification of Trichoderma spp. in Mushroom Farms in Serbia

Disease with symptoms resembling green mould was noted in mushroom facilities in Serbia. The aims of this study were to isolate and identify the causal agents of green mould, examine variation of the pathogen as evidenced by morphology of its colonies, using molecular biology techniques and their pathogenic characteristics. Pathogenicity assay was performed on harvested basidiomes of A. bisporus treated with a 3 X 106 conidia/mL of spore suspensions of all tested Trichoderma spp. isolates. Identification of Trichoderma spp. was based on colonial morphology, optimal growth temperature, odor, conidial morphology and conidia length and width. Identification was confirmed using molecular techiques using sequence analyses of ITS1/4 amplicon. Diseased fruiting bodies of A. bisporus with symptoms resembling green mold disease were observed in 14 mushroom units during 2006-2010. Nectrotic lesions and brown, rust coloured spots were observed on caps of fruiting bodies of eleven A. bisporus and one Lentinus edodes mushroom farms from Serbia and one A. bisporus farm from Bosnia and Hezegovina. Samples of compost with large dark areas of dense green sporulation were collected from one mushroom farm in Serbia. Pathogenicity assay on mushroom pilei showed that each of the tested isolates had high virulence level for A. bispours. The isolates were classified according their morphological characteristics as:T. atroviride, T. koningii, T. virens, T. aggressivum f. sp. aggressivum and T. harzianum. The phylogenetic analyses on ITS1/4 amplicons confirmed the differentiation obtained with classical mycological methods.
