This study addresses controls for an assay of bacterialrnvirulence that has been optimized for space flightrnstudies. Caenorhabditis elegans (C. elegans) wormsrningest microorganisms, but are also killed by virulentrnbacteria. Virulence is assessed by the number of bacteriarnsurviving in co-culture with C. elegans, as measuredrnby optical density at 620 nm. Co-cultures ofrnMethicillin-resistant Staphylococcus aureus (MRSA)rnwith C. elegans have a higher OD620 than MRSArngrown alone, which could reflect debris from deadrnworms and/or enhanced growth of the MRSA in responsernto worm-derived factors. The use of mediarnconditioned by pre-incubation with worms demonstratedrnthe presence of temperature-stable factors thatrnchange MRSA growth in a strain-dependent manner.rnSome sources of deionized water contain an undefinedrnantibacterial activity present in conditioned, butrnnot fresh untreated media.
