Cis-acting sequences can act as important signals affecting the expression of genes in eukaryotes. I have analyzed DNA sequence signals of two types of genes from Bombyx mori that are transcribed by RNA polymerase III, the polymerase responsible for the expression of tRNA, 5S RNA, and other small RNA gene sequences.;The tRNA('Ala) control region is large relative to control regions previously identified for tRNA genes in other organisms. To understand this discrepancy, in this work I have studied the effect of variation of assay conditions on the extent of sequences that appear to be important for transcriptional control. These experiments show that certain assay parameters have a determinative effect on the apparent size of the tRNA('Ala) control region; certain conditions can obscure the importance of portions of tRNA('Ala) control sequences. My results suggest that previous transcriptional analysis of tRNA genes may not have identified all sequences that contribute to the success of the reaction.;Second, I have studied the transcription of a previously undescribed gene sequence, BmX, by the silkworm RNA polymerase III. I discovered the BmX template, and have characterized the family of repetitive DNA sequences (20,000 copies/genome) to which it belongs. I have analyzed in detail a member of the BmX gene family, identifying transcriptional initiation and termination sites. To identify the transcriptional control region of the BmX gene, mutant derivatives of the BmX sequence were analyzed in in vitro transcription experiments. Like the tRNA('Ala) gene control region, the BmX control region is large, including 5' flanking, coding, and 3' flanking sequences. BmX gene sequences also bind the limiting transcription factor(s) in silkworm crude extracts.;First, I have characterized sequences that contribute to the expression of a tRNA('Ala) gene in in vitro transcription experiments. The tRNA('Ala) control region defined in this work is very large, including (TURN)30 bp of 5' flanking DNA, coding sequences, and (TURN)50 bp of 3' flanking DNA. A subset of these tRNA('Ala) control sequences are essential for binding a limiting transcription factor(s) found in crude cell extracts.
