Experiments were conducted to investigate whether androgen-inhibition of tonic follicle-stimulating hormone (FSH) secretion in the hamster, Mesocricetus auratus, undergoes sexual differentiation during early postnatal life. Physiological concentrations of serum testosterone (T) (2.8 (+OR-) 0.3 ng/ml) inhibited 66-75% of the serum FSH response and 100% of the LH response to orchidectomy. Higher levels of serum T (> 4.7 (+OR-) 0.5 ng/ml) suppressed serum FSH below the physiological range (229 (+OR-) 18 ng/ml). Equivalent serum T concentrations prevented the serum LH response to ovariectomy, but serum FSH levels were only partially (30%) suppressed and remained greater than 500 ng/ml regardless of circulating titers of T. Males showed parallel suppression of serum and pituitary FSH in response to T; whereas, females exhibited elevated pituitary FSH as serum T and FSH levels increased.;Orchidectomy at birth (day-0) resulted in reduced sensitivity to T-inhibition of FSH secretion during adulthood. Testosterone propionate (TP) injected on days 0 and 1 prevented this desensitization in neonatally orchidectomized males, and rendered females more sensitive to T-inhibition of tonic FSH secretion during adulthood. Estradiol benzoate (EB) injected on days 0 and 1 did not influence tonic FSH responses to T during adulthood, although these females were anovulatory and had masculinized genitalia.;In adult hamsters, estradiol-17(beta) (E(,2)) and 5(alpha)-dihydrotestosterone (DHT) prevented serum FSH responses to orchidectomy. E(,2) prevented the serum FSH response to ovariectomy, but DHT was ineffective in this regard. Neonatal orchidectomy rendered adults unresponsive to DHT-inhibition of FSH secretion. Neonatal TP-treatment caused 2 of 7 females to respond to DHT-inhibition of tonic FSH secretion during adulthood.;These findings suggest that (a) partial inhibition of tonic FSH secretion in T-treated females results from estrogenic metabolities of T, (b) the capacity for androgenic (i.e. DHT) inhibition of tonic FSH secretion develops as a result of testicular androgen action during early postnatal life, and (c) this early organizational action of androgen is not mediated by aromatization to E(,2). Thus, mechanisms that mediate androgen-inhibition of tonic FSH release undergo sexual differentiation during neonatal development.
