小麦逆境胁迫相关基因 TaCBL7的克隆、生物信息学及表达特性分析

         

摘要

从小麦抗性相关EST数据库中筛选并获得一条753 bp的核苷酸序列,该序列包含了一个642 bp的开放阅读框,编码一个由213个氨基酸残基组成的蛋白,氨基酸同源性分析发现,该蛋白含有保守的钙结合结构域EF-hand ,在其N端含有豆蔻酰化位点MGXXXS/T,与OsCBL7高度同源,属于小麦CBL家族,命名为TaCBL7。实时荧光定量PCR分析表明,TaCBL7基因在苗期的根、茎及叶子,成熟期的根、茎、叶子及种子中均表达,但表达存在差异;该基因表达受盐、干旱、低温、ABA及条锈病诱导调控,表达分析结果表明,TaCBL7基因在小麦发育时间和组织空间上存在表达特异性(即时空表达特异性),同时参与了小麦生物与非生物胁迫的响应。推测TaCBL7基因编码的蛋白在小麦生长发育及胁迫过程中具有重要功能,为进一步研究小麦TaCBL7基因在生长发育和逆境响应过程中的功能,以及利用TaCBL7基因进行小麦分子育种奠定基础。%The cDNA sequence of TaCBL7 were isolated and analyzed from wheat EST database .The results showed that TaCBL7 contained a 642 bp ORF encoding 213 amino acids .Using the methods of bioinformatics to predict and analyze TaCBL7 ,the protein has the high level homology with OsCBL 7 and highly conserved domain of EF-hand and N-myristoylation .The results of Real-time PCR reveal that the expression of TaCBL7 has expression in leaf,stem,seed and root both seedling and mature stage .In addition,the expression of TaCBL7 was induced by drought,salt,low-temperature,ABA and yellow rust.Those results indicate that TaCBL7 has specific expression of temporal and spatial ,and the expression was induced by biotic and abiotic stresses .It is speculated that TaCBL7 could play a role not only in growth and development conditions but also in stress of wheat .This investigation laid a foundation for functional dissection of TaCBL7 and wheat molecular breeding with TaCBL7 in the future .
