淋病奈瑟菌共同抗原表位在淋病快速检测中的临床应用价值

     

摘要

Objective To study the applied value of common epitope of gonococcus in rapid detection of gonorrhea. Methods A polypeptide(WSWLF) was synthesized according to the amino acid sequence of common epitope in the outer membrane protein PIB of different types of gonococi. The polypeptide was coupled with Keyhole limpet hemocyanin(KLH)to construct complete antigen WSWLF-KLH to immune animals and the antiserum was harvested. FLISA and Western blot were used to detect the immunogenicity and specificity. The antiserum was used for detecting the blood serum of 169 cases of the clinically diagnosed gonorrhea and compared with the traditional gold standard method. Results FLISA revealed that WSWIF-KLH had immunogenicity. Strong humoral immune reaction appeared in immunized animals. The valences of rabbit serum antibody and guinea pig antibody could reach 1 : 3 200 and 1 : 25 600 respectively. The antiserum could specifically bind with clinical strains of gonococci , but it had no cross reaction with staphylococci , streptococcus , colibacillus or eumycete etc. Western blot showed the antiserum could identify gonococcus lysate and produce specific strap, with relative molecular mass of 37 300. The similarity of detection rate between this method and traditional immune colloidal gold technique was 93. 49 %. Conclusion Common epitope of gonococcus has immunogenicity. Immune serum can bind with most gonococci. The common epitope of gonococcus has great clinical value in rapid detection of gonorrhea.%目的 研究淋病奈瑟菌共同抗原表位在淋病快速检测中的临床应用价值.方法 根据各型淋病奈瑟菌外膜蛋白PIB上共同抗原表位的氨基酸序列合成一段多肽(WSWLF),并将其与钥孔血蓝蛋白(KLH)偶联构成完全抗原WSWLF-KLH,免疫动物,收集抗血清,ELISA和Western blot检测WSWLF-KLH免疫原性和特异性,并用于临床已确诊的169例淋病患者血清的检测及与传统金标准方法进行对比.结果 实验室ELISA检测表明,WSWLF-KLH具有免疫原性,免疫动物能刺激机体产生较强的体液免疫反应,兔血清抗体和豚鼠血清抗体效价最高分别达到1∶3 200和1∶25 600;抗血清能与淋病奈瑟菌临床株特异性结合,与葡萄球菌、链球菌、大肠埃希菌和真菌等其它微生物之间没有交叉反应.Western blot结果显示,抗血清能识别淋病奈瑟菌裂解物,产生特异性条带,相对分子质量为37 300.临床实验结果显示,抗血清的淋病检出率与传统的金标准方法结果相似率为93.49%.结论 淋病奈瑟菌共同抗原表位具有免疫原性,免疫血清能与大多数淋病奈瑟菌结合,该抗原表位作为快速检测试剂有较高的临床应用价值.
