ω-3多不饱和脂肪酸对严重烫伤大鼠肝脏及炎症反应的影响

         

摘要

目的 观察ω-3 多不饱和脂肪酸(ω-3 PUFA)对严重烫伤大鼠肝脏及炎症反应的影响,并就其对肝脏保护作用的机制进行探讨.方法 72只SD大鼠随机分为3组:假烫组8只、对照组(中长链脂肪酸组)32只和治疗组(ω-3 PUFA组)32只.采用大鼠30%总体表面积(TBSA)Ⅲ度烫伤模型.对照组每天按时经尾静脉注射与治疗组相同热量的中长链脂肪酸;治疗组经尾静脉注射ω-3 PUFA 2 ml/(kg·d);假烫组不予任何处置.观察假烫组、对照组和治疗组烫伤后1、4、7、10 d血清内毒素(ET)、前列腺素E2(PGE2)的含量;免疫组化检测肝组织核因子-κB(NF-κB)p65的活性水平;RT-PCR检测肝组织肿瘤坏死因子α(TNF-α) mRNA的表达,各个时间点均处死8只大鼠.结果 ① 在伤后所有时间点,对照组、治疗组血清ET、PGE2水平明显高于假烫组(P<0.01);治疗组血清ET、PGE2水平比对照组降低,伤后4、7、10 d与对照组比较,差异有统计学意义(P<0.05).② 伤后对照组、治疗组肝组织NF-κBp65表达比假烫组增强;在伤后4、7 d,与对照组比较,治疗组肝组织NF-κBp65表达减弱(P<0.05).③ 对照组、治疗组在烫伤后各时间点,肝组织TNF-α mRNA表达较假烫组明显升高(P<0.01),与对照组比较,在烫伤后4、7 d,治疗组肝组织TNF-α mRNA的表达明显降低(P<0.05).结论 ① ω-3 PUFA能降低烫伤后血清ET、PGE2的含量,减轻肝脏组织NF-κBp65和TNF-α mRNA的表达.② ω-3 PUFA减轻严重烫伤后肝组织的损伤.%Objective To study the influence of ω-3 polyunsaturated fatty acids (ω-3 PUFA ) on liver and inflammatory reaction in serious scalded rats and explore its mechanism. Methods A total of 72 SD rats with 30% total body surface area ( TBSA ) third-degree scalded were optional separated into 3 groups: the phony scald group 8, the control group ( middle-long-chain fatty acids group ) 32 and treatment group ( ω-3 PUFA group )32. Rats in the control group were injected middle-long-chain fatty acids with equivalent calories by tail vein, and rats in the treatment group were injected ω-3 PUFA 2 ml/( kg·d ) by tail vein on time for everyday. The phony scald group,the control group and treatment group 1 ,4,7,10 d were measured respectively the levels of serum endotoxin ( ET ), prostaglandinE2( PGE2 )and expression of nuclear factor-kappa B( NF-ΚB )p65 in liver tissue was detected by im-munohistochemistry staining. Expression of liver tissue TNF-α mRNA was measured by RT-PCR technology. Each subgroup had 8 rats. Results ①Serum ET, PGE2 were obvious higher than the phony scald group at each time point in the control group and treatment group ( P <0. 01 ). The levels in the treatment group were lower than in the control group, and were significantly different in 4, 7,10 d after scalded ( P < 0. 01 ). ②Compared with the phony scald group, the expression of liver tissue NF-κBp65 was increased in the control group and treatment group; Compared with the control group, the activity of NF-κBp65 was significantly decreased after scalded 4, 7 d in the treatment group ( P <0. 05 ). ③The control group and treatment group at all time points after scalded the expression of liver tissue TNF-α mRNA was significantly higher than the phony scald group( P <0. 01 ); Compared with the control group, after scalded 4, 7d, the expression of liver tissue TNF-α mRNA was significantly reduced in the treatment group( P <0. 05 ). Conclusion ① ω-3 PUFA can reduce the levels of serum ET, PGE2, relieve the expression of liver tissue NF-κBp65 and TNF-α mRNA. ②ω-3 PUFA can alleviate the damage of liver tissue after scald.
