血小板冻干基础保护剂及复水液的比较分析

         

摘要

Objective To explore the basic protectant and rehydrating solution in the suitable glycerol content of the platelets freeze -dried.Method Different concentrations of glycerol plasma/glycerol saline/glycerol water were used for preservation of the platelets freeze-dried agent ,and water/saline was used for the rehydrating solution of the freeze-dried platelets .Different osmolality value of the platelets freeze-dried basic protectant was compared ,moreover ,MPV,PDW and recovery of the freeze-dried platelets after rehydration were compared . Result There were different osmolality valuesintra-group and inter-group of the various platelets freeze-dried basic protectant .As the intra-group glycerol concentrations was increased , the osmolality values of platelets freeze-dried basic protectant in group A , B and C were in-creased.There was part of the MPV and PDW difference intra-group and inter-group of the various platelets freeze-dried basic protectant after rehydrating platelets freeze-dried.The MPV of the group A1 platelets after rehydrating was the maximum .The PDW of the group C2 platelets after rehydrating was the highest .There was difference recovery intra-group and inter-group of the various platelets freeze-dried basic pro-tectant after rehydrating platelets freeze-dried.The recovery of the group A 1 platelets after rehydrating was the optimum .Particularly 7%glyc-erol plasma protectant for preservation of the platelets freeze-dried agent and water for the rehydrating solution of the freeze-dried platelets ,the recovery of the platelets after rehydrating was (85.5 ±13.7)%.Conclusion The 7% glycerol plasma protectant for preservation of the platelets freeze-dried agent and water for the rehydrating solution of the freeze-dried platelets is the most appropriate .The comparative analysis provides fundamental information for the study of added protection agents in basic protectant and the platelets function activity .%目的:探讨适宜甘油含量血小板冻干基础保护剂和复水液。方法应用不同浓度甘油血浆/甘油生理盐水/甘油纯水作血小板冻干基础保护剂,纯水/生理盐水作复水液,比较不同血小板冻干基础保护剂渗透压值,同时比较冻干血小板复水后平均血小板体积( MPV)、血小板体积分布宽度( PDW)和回收率的差异。结果不同血小板冻干基础保护剂组间、组内渗透压值存在差异,血小板冻干基础保护剂A组、B组和C组组内随着甘油含量增高基础保护剂的渗透压值增加。不同血小板冻干基础保护剂组间、组内复水后血小板的MPV和PDW值存在差别,A1组复水后血小板的MPV值最大,C2组复水后血小板的PDW最高。不同血小板冻干基础保护剂组间、组内复水后血小板回收率存在差异,A1组冻干血小板复水后回收率最佳,尤以7%甘油血浆基础保护剂纯水复水回收率最高,达(85.5±13.7)%。结论7%甘油血浆基础保护剂和纯水为适宜的血小板冻干基础保护剂和复水液,此为开展冻干血小板基础保护剂中添加保护剂及血小板功能活性研究奠定了基础。
