黑荞麦中花青素的提取及含量测定

         

摘要

We established a method of microwave extraction of anthocyanins in black buckwheat and content determination by HPLC.With 60%(v/v)ethanol as extraction solvent,solid-liquid ratio of 1∶15,temperature of 60℃,microwave extrac-tion for 15 min,chromatographic column Shim-pack-vp-ODS (150 mm × 4.6 mm,5 μm),mobile phase of methanol (0.02 mol/L)-potassium dihydrogen phosphate (with phosphoric acid to adjust pH = 3.0)(80∶20),the flow rate was 1.0 ml/min,and sample quantity was 10μl.The results showed that:in the range of 0.04-0.28 mg/ml,the regression equa-tion had good linear relationship,precision was 0.76%,the recovery was 99.0%-100.2% .The method was simple and rapid, good reproducibility and quantitative accuracy,high accuracy and precision,as a result,suitable for determination the content of anthocyanin in black buckwheat.%建立了一种用微波提取黑荞麦中花青素,并用高效液相色谱法测定其含量的方法。以体积分数60%乙醇为萃取溶剂、料液比为1∶15、温度为60℃,微波萃取15 min;以Shim-pack-vp-ODS(150 mm×4.6 mm,5μm)色谱柱,甲醇(0.02 mol/L )-磷酸二氢钾(用磷酸调节pH=3.0)(80∶20)为流动相,流速为1.0 ml/min,进样量为10μl进行检测。黑荞麦中花青素含量在0.04~0.28 mg/ml,回归方程具有良好的线性关系,精密度为RSD=0.76%,回收率为99.0%~100.2%。该方法简便快速,结果重现性好,定量准确,准确度、精密度高,适用于黑荞麦中花青素的含量测定。
