The interaction of antitumor ruthenium methylimidazole complexes[Ru(MeIm)4(L)]2+(L=iip,Ru1;L=tip,Ru2)with telomere DNA was studied by UV-Visible spectra,circular dichroism (CD )spectroscopy,fluorescence resonance energy transfer (FRET)assay and so on. The experimental results showed that the title complexes could bind to telomere DNA more tightly and in-duce the stabilization of quadruplex-DNA. Moreover,the ability of complex Ru2 to stabilize quadruplex-DNA was stronger than that of complex Ru1.%采用紫外-可见吸收光谱、圆二色谱、荧光共振能量转移等实验研究了具有抗肿瘤活性的钌甲基咪唑配合物[Ru(MeIm)4(L)]2+(L=iip,Ru1;L=tip,Ru2)与端粒DNA:HTG21(AG3(T2AG3)3)的相互作用。结果表明钌配合物与端粒DNA之间存在较强的亲合力,能够诱使HTG21形成G-四链体结构并稳定该结构,且Ru2的作用大于Ru1。
