Currently thermal tissue damage is mainly evaluated with histological examination which requires the removal of a section of tissue.There is no ideal method to detect molecular changes after thermal damage tissue. The research designed a new method to evaluate thermal tissue damagein situ using native fluorescence spectroscopic imaging under 340 nm excitation. Native fluorescence spectroscopic imaging was performed on laser irradiated bovine tendon tissue. The fluorescence imaging of the tendon tissue were confirmed and compared with the image of the laser-irradiated tendon tissue with the histological method of picrosirius red F3BA stain. The results of the experiment indicate that the changes in native lfuorescence from collagen on the laser-irradiated tissue could be observed on the thermal damage region of the tissue.%目前组织热损伤主要依靠离体切片组织学进行评价,尚没有一种较理想的评价方法。本研究设计了一种利用340 nm波长的自体荧光成像技术评估活体组织热损伤的方法,并采用该方法对激光照射后的牛腱组织进行了荧光成像,将荧光成像后的牛腱组织样本与日光灯下成像后的牛腱组织样本通过天狼星红F3BA染色的组织学方法进行了验证和比较。实验结果显示,样本中受激光照射区域的胶原蛋白的自体荧光发生了明显改变,说明该方法能够用于评价组织热损伤。
