目的:初步研究罗汉松种子提取物对人胃癌细胞MGC-803的抑制作用。方法采用噻唑蓝(MTT)法测定不同浓度梯度的罗汉松种子提取物作用人胃癌MGC-803细胞24,48,72 h的增殖抑制率。采用RT-PCR检测MGC-803细胞凋亡相关基因bcl-2和p53 mRNA的表达。结果不同质量浓度罗汉松种子提取物对MGC-803细胞均有增殖抑制作用,且随着质量浓度的增加和作用时间的延长,抑制率呈上升趋势。RT-PCR结果显示,不同质量浓度罗汉松种子提取物作用于MGC-803细胞,bcl-2 mRNA水平随质量浓度的增加而明显下降,而p53 mRNA水平则随质量浓度的增加而显著增强。结论罗汉松种子提取物对人胃癌细胞MGC-803有明显的体外抑制作用,其抑制作用与药物的质量浓度和作用时间呈正相关,可能与其调控相关凋亡基因的表达有关。%Objective To preliminarily study the inhibition effect of the podocarpus seed extract on human gastric cancer cell MGC-803. Methods The methyl thiazolyl tetrazolium ( MTT ) assay was adopted to detect the the 24, 48, 72 h proliferation inhibition rate of human gastric cancer MGC-803 cells caused by different concentrations gradient of the podocarpus seed extract. RT-PCR was adopted to detect the expression of MGC-803 apoptosis-related genes P53 and BcL-2 mRNA. Results Different concentrations of podocarpus seed extract had the proliferation inhibiting effect on MGC-803 cells, moreover as the increase of podocarpus seed extract concentration and the extension of the treatment time, the inhibition rate showed the upward trend. The RT-PCR results showed that in different concentrations of podocarpus seed extract acting on the MGC-803 cells, the expression level of BcL-2 mRNA was decreased with the concentration increase, while the expression level of P53 mRNA was significantly enhanced with the concentration increase. Conclusion Podocarpus seed extract has significantly inhibiting effect in vitro on human gastric cancer cell MGC-803, and its inhibition effect is positively correlated with the drug concentration and the action time, which may be related to the regulation of the apoptosis-related genes expression.
