miR-let-7基因家族在金华猪和长白猪中的组织表达分析

         

摘要

miR-let-7基因广泛参与动物组织器官的发育调控,其表达具有组织特异性.本研究选用70 d的金华猪和长白猪公猪(各3头),采用实时荧光定量PCR(RT-PCR)技术检测了let-7基因家族6个成员(let-7a、let-7c、let-7d-3p、let-7c、let-7f、let-7g)在心脏、肝、脾、肺、肾、小肠中的表达量,分析它们在组织间及品种间的表达差异,重点分析了在小肠中的表达差异.结果表明:let-7基因家族在长白猪肝组织中表达量显著高于其他组织(P<0.05),在金华猪小肠和心脏中显著高于其他组织(P<0.05);在小肠中,金华猪let-7a和let-7c基因表达量极显著高于其他成员(P<0.01),长白猪let-7a基因表达量极显著高于其他成员(P<0.01),let-7在小肠的表达存在品种间差异,金华猪的let-7a和let-7d-3p基因表达量极显著高于长白猪(P<0.01),let-7c基因显著高于长白猪(P<0.05),而let-7e基因却显著低于长白猪(P<0.05).本研究结果为探究miR-let-7基因在金华猪和长白猪生长和脂肪形成方面的分子作用机制提供了参考.%miR-let-7 gene family is a tissue-specific expressed and widely involved in the regulation and the development of animal tissues and organs.In our study,three of each Jinhua and Landrace pigs at 70 days old were selected to detected the expression of let-7 gene family (let-7a,let-7c,let-7d-3p,let-7e,let-7f,,let-7g) in six tissues (heart,liver,spleen,lung,kidney,small intestine).The differential expression of let-7 genes were analyzed in various tissues and especially in the small intestine for the Jinhua and Landrace pigs.The results showed that the expression level of let-7 genes were abundantly expressed in liver of landrace (P <0.05) while the expression of let-7 genes in the heart and small intestine of Jinhua pig were greater than other tissues(P <0.05).In the small intestine of Jinhua pig,the expression levels of let-7a and let-7c genes were significantly higher than other members of the let-7 gene family;the expression level of let-7a gene was observably higher than other members in Landrace.The expression levels of let-7a and let-7d-3p gene in small intestine of Jinhua was significantly higherthan those of Landrace (P<0.01).The expression of let-7c gene was higher than that of Landrace (P <0.05).But the expression of let-7e gene was lower than that of Landrace (P <0.05).The expression ofmiRlet-7 gene family was studied in this paper,it provides useful clues for further study of molecular mechanism ofmiR-let-7 on the growth and fat formation for Jinhua and Landrace.
