目的:探讨基于色谱/质谱联用(LC/MS)及模式识别为主的代谢组学技术及其方法,研究慢性心力衰竭肾阳虚证的代谢模式及相应代谢网络的变化.方法:收集正常人以及临床常见慢性心力衰竭肾阳虚证与气阴两虚证病例的尿样,以LC/MS为代谢组学研究方法,检测分析各组间尿样中代谢模式的差异.结果:代谢组学研究表明,与正常组比较慢性心衰组有21种内源性化合物含量与正常组有明显不同;与气阴两虚组比较,慢性心力衰竭肾阳虚组有16种内源性化合物含量与气阴两虚组有明显不同.结论:以代谢模式改变为依据的代谢组学研究方法,通过代谢通路成功地将不同证型加以区分,说明代谢组学技术及其方法研究中医学证本质的思路及方法可行,从代谢模式的角度阐述中医证本质是中医现代化可实践和深化的技术和方法学体系.%Objective:To investigate kidney yang deficiency metabolic patterns and metabolic networks changes by metabolomics which based on GC/MS and pattern recognition.It is to construct a methodology to suitable for TCM's syndrome theories.Methods:Collecting urine samples of chronic heart failure patients who appear kidney yang deficiency syndrome and Qi deficiency and Yin deficiency syndrome.Then their different metabolic patterns is to be distinguished metabolic profiling changes.72 patients were divide into 2 groups:38 chronic heart failure cases in the kidney deficiency syndrome group,24 chronic heart failure cases in the Qi deficiency and Yin deficiency syndrome group,48 normal people to be treated as control group.And collecting the first time urine samples after hospitalization is to be collected and applied to study metabolic pattern change of control group,kidney Yang deficiency and Qi deficiency and Yin deficiency syndrome.Results:The metabolomics indexes showed chronic heart failure and control group's metabolomic pattern graph distribute different area,at the same time chronic heart failure kidney yang deficiency group and Qi deficiency and Yin deficiency syndrome group metabolomic pattern graph also distribute different area.Conclusion:The metabolomics is showed chronic heart failure and normal people can be distinguished by LC/MS.Meanwhile chronic heart failure and chronic kidney failure disease's kidney yang deficiency and non-kidney yang deficiency can be distinguished by LC/MS.They discriminate kidney yang deficiency and non-kidney yang deficiency different metabolomic pattern.The results certificate metabolomics technology can be applied in the investigation of TCM syndrome.This technology should be expanded and strengthened.
