电针对抑郁模型大鼠海马脑源性神经营养因子基因表达的影响

         

摘要

目的:探讨电针治疗抑郁症的部分作用机制.方法:大鼠随机分为正常组、模型组和电针组,用慢性不可预见性轻度刺激结合孤养的方法造模,造模同时电针组选取"百会"、"印堂"穴进行治疗.于实验前1d、实验第7、14、21天进行开野实验及精水消耗实验,用原位杂交法检测大鼠海马组织脑源性神经管养因子(BDNF)mRNA的表达.结果:电针组的水平运动与垂直运动次数始终高于模型组,第21天电针组相对糖水消耗量高于模型组,电针组的BDNF mRNA杂交阳性信号明显高于模型组(均P<0.05).结论:电针"百会"、"印堂"穴能够干预抑郁症状的出现,与其提高抑郁模型大鼠海马区BDNF mRNA的表达量有关.%Objective; To explore the underlying mechanism of EA in improving depression. Methods; Thirty rats were evenly randomized into control, model and EA groups. Depression model was established by using chronic unpredictable mild stress stimulation combined with lonely raising for 21 days. EA was applied to "Baihui" ( GV 20) and "Yintang" ( EX-HN 3 ) , once daily, continuously for 21 days. Open-field test, relative volume of sucrose-intake were recorded one day before experiments,and on the 7th, 14th and 21st day during experiments respectively. The expression of BDNF mRNA in the hippocampus was determined by nucleic acid in situ hybridization. Results ; In comparison with model group,the rats' crossing and rearing movement times from 7th to 21st day, relative volume of sucrose-intake on the 21st day increased considerably in EA group ( P < 0. 05 ) . The positive hybridization products of EA group were obviously higher than those of model group (P < 0. 05 ). Conclusion; EA of "Baihui" ( GV 20) and "Yintang" ( EX-HN 3) can upregulate the expression of BDNF mRNA in hippocampus in depression rats,which may contribute to its effect in relieving depression.
