Response surface analysis was used to optimize for α⁃pinene production. Initially, MD beef extract powder was determined to be the best nitrogen source for α⁃pinene production, then Plackett⁃Burman ( P⁃B ) experiment was used to evaluate the eight nutritional variables to screen medium component that significantly influence the α⁃pinene production and their optimal levels. Results showed that beef extract powder,pH and citric acid were significant factors. Then the path of steepest ascent was used to approach the optimal region of the above three factors. In the third step,the concentration of beef extract powder,pH and citric acid was further optimized using central composite designs and response surface analysis. As the result,the concentration ofα⁃pinene increased from 1�710 to 7�053 mg/L. With the optimized medium,the yield of α⁃pinene increased about 3⁃fold.%利用响应面法对α蒎烯发酵培养基进行优化。先利用单因素实验法筛选出MD牛肉粉为最有利于发酵产α蒎烯的N源。此后用Plackett Burman ( P B)实验方法对相关影响因素效应进行评估,筛选出有显著效应的3个因素:柠檬酸用量、牛肉粉用量以及培养基初始pH。然后用最陡爬坡实验逼近以上3因素最优水平。最后由中心组和实验设计法及响应曲面分析法确定主要因素的最佳条件。在优化后的培养条件下,发现α蒎烯产量从1�710 mg/L提升到7�053 mg/L,发现产量提高了3倍左右。
