干细胞因子对高糖环境下豚鼠膀胱Cajal样间质细胞c-kit mRNA及蛋白表达的影响

         

摘要

目的 观察干细胞因子(stem cell factor,SCF)对高糖环境下豚鼠膀胱Cajal样间质细胞c-kit mRNA及蛋白的表达影响.方法 胶原酶消化法原代分离培养豚鼠膀胱Cajal样间质细胞,培养24 h后分别加入葡萄糖浓度为5 mmol/L(正常对照组)、15 mmol/L(高糖组)培养基培养72 h后,高糖组分为高糖对照组、SCF干预组(50 ng/ml、100 ng/ml),分别继续培养24 h、72 h,应用RT-PCR、激光扫描共聚焦显微镜(LSCM)分别检测膀胱Cajal样间质细胞c-kit mRNA和蛋白表达变化.结果 高糖对照组较正常对照组c-kit mRNA及蛋白表达均明显下降.24 h组中,SCF50 ng/ml较高糖对照组c-kit mRNA(P>0.05)和蛋白表达(P>0.05)差异不大,SCF100 ng/ml较高糖对照组c-kit mRNA(P<0.05)和蛋白表达(P<0.05)升高;72 h组中,SCF50 ng/ml组、SCF100 ng/ml组较高糖对照组c-kit mRNA(P<0.01)和蛋白表达均明显升高(P<0.01).结论 高浓度SCF可促进已发生形态、机能变化的离体膀胱Cajal样间质细胞c-kit表达上调.%Purpose To observe the stem cell factor ( SCF ) intervention on interstitial cells of Cajal-like cells ( ICCs-like ) c-kit mRNA and protein expression in the bladder of guinea pig in the high glucose environment. Methods The ICCs-like cells were isolated from the bladder of guinea pig by collagenase digestion and cultured for 24 h, then medium of 5 mmol/L ( normal control group ), 15mmol/L ( high glucose ) glucose concentration was added and continued to culture for 72 h. The high glucose group was divided into high glucose control group and SCF intervention group ( 50 ng/ml,l00 ng/ml ) and cultured for 24 h and 72 h. Last mRNA expression of c-kit was detected by reverse transcription polymerase chain reaction ( RT-PCR ) and protein expression of c-kit was tested and analyzed by laser scanning confocal microscope. Results Compared to control group, c-kit mRNA and protein expression levels of high glucose were significantly decreased. In 24 h group, c-kit mRNA and protein expression in SCF 50 ng/ml group did not increased significantly than high glucose control group( P > 0. 05 ). However, c-kit mRNA and protein expression in SCF 100 ng/ml group were higher than that in high glucose control group( P < 0. 05 ). In 72 h group , c-kit mRNA and protein expression in SCF50 ng/ml and 100ng/ml group were higher than that in high glucose control group( P < 0. 01 ). Conclusion High concentrations of SCF mav increase the expression of c-kit in interstitial cells of Cajal which have changed in shape and function.
