Objective:To study the expression level of CD74 receptor of macrophage migration inhibitory factor(MIF) in cell lines and tissues of human breast cancer and its correlation with prognosis. Methods: The expression levels of CD74 mRNA and protein were detected by fluorescent quantitative polymerase chain reactionCFQ - PCR) and Western blotting in breast cancer cell lines MCF - 7 and MDA - MB- 231 which were different in the potential of metastasis;the expression of CD74 in 89 cases of breast cancer was detected by immunohistochemistry. The relationship between CD74 expression and overall survival (OS) and disease - free survival (DFS) of the breast cancer patients was analyzed. Results: The expression level of CD74 in high metastatic potential cell strain MDA - MB- 231 was higher than that in the low metastatic potential cell strain MCF - 7(P<0. 001 ). CD74 expression was an independent prognostic factor of OS(P = 0. 001 )and DFS(P<0. 001). Conclusions: The expression level of CD74 is an independent prognostic factor in breast cancer.%目的:研究巨噬细胞迁移抑制因子受体CD74在乳腺癌细胞株MCF-7和MDA-MB-231 以及人乳腺癌组织中的表达及其与乳腺癌患者预后的关系.方法:应用荧光定量聚合酶链式反应(fluorescent quantitative polymerase chain reaction,FQ-PCR)和蛋白质印迹(Western blotting)方法分别检测不同转移潜能乳腺癌细胞株MCF-7和MDA-MB-231 CD74的mRNA和蛋白表达量,并比较CD74在两者间的表达差异;应用免疫组织化学检测89例乳腺癌组织中CD74的表达情况,分析CD74与乳腺癌患者总生存率和无瘤生存率的关系.结果:高转移潜能乳腺癌细胞株MDA-MB-231中CD74表达显著高于低转移潜能细胞株MCF-7(P<0.001).CD74表达水平是乳腺癌总生存率(P=0.001)和无瘤生存率(P<0.001)的独立预测因素.结论:CD74的表达水平与乳腺癌患者预后呈负相关.
