摘要

AIM: To observe the Ca metabolism in cultured rat Calvaria with various combinations of PGE2,TNF-α and IL-1β. METHODS:Microelement examination was carried out on the conditioned media. RESULTS: ①The combination of PGE2 and IL-1β had Ca absorptive activity at 0.01ng/ml level. Which is decreased with increasing of the concentrations. ②the Combinations of PGE2 and TNF-α had Ca absorptive activity at 0.01~0.01ng/ml level. There are marked releasing activity of Ca at 1.0ng/ml level.③The combinations of IL-1β and TNF-α had Ca absorptive activity at 0.01ng/ml level. There are no marked absorptive or releasing activity of Ca at 1.0ng/ml level.④The combinations of PGE2、IL-1β and TNF-α had Ca absorptive activity, which is rised with the increasing of the concentrations at 0.01~1.0ng/ml Levels. CONCLUSION: The various combinations of PGE2, TNF-α and IL-1β had interactions, antagonistic effects, synergism on the Ca metabolism of cultured rat calvaria in vitro. PGE2 mainly shows being the synergie factor%目的:观察不同浓度组合的前列腺素E2(PGE2)、肿瘤坏死因子α(TNF-2)、白细胞介素1β(IL-1β)对培养骨组织钙代谢的作用。方法:原子吸收分光光度计检测骨组织培养上清液中Ca2+浓度。结果:①PGE2+IL-1β:在0.01~1.0ng/ml浓度组合时,有促进钙吸收的作用,此作用随组合浓度的加大而减弱。②PGE2+TNF-α:0.01~0.1ng/ml浓度时,有钙吸收作用,而在1.0ng/ml浓度组合时,有明显的钙释放作用。③IL-1β+TNF-α:0.01ng/ml浓度组合时,有钙吸收作用。0.1ng/ml浓度组合时,钙吸收、释放作用不明显。1.0ng/ml浓度组合时,强刺激脱钙。④PGE2+TNF-α+IL-1β:0.01~1.0ng/ml浓度组合时显示钙吸收作用,且随剂量增加而增加。结论:3种细胞因子不同组合后,有交互作用、拮抗作用和协同作用。PGE2主要表现为协同因子而发挥作用。
