目的 探讨miR-145在宫颈上皮内瘤变III级(CINIII级)和宫颈鳞癌患者组织中的表达情况及与宫颈鳞癌发生之间的关系.方法 2010年11月-2011年8月在内蒙古医科大学第一附属医院提取49例宫颈鳞癌组织(宫颈癌组)、27例宫颈上皮内瘤变III级和2例原位癌(CINIII级组)、11例宫颈良性病变患者(健康对照组)组织中的miRNA,用实时定量PCR的方法分析miR-145在各组患者组织中的表达情况.结果 (1)宫颈鳞癌患者组织中miR-145的表达较健康对照组、CINIII级组降低(P＜0.05),miRNA-145在宫颈癌组织中的表达是CINIII级组的0.579倍,是健康对照组的0.521倍;(2)miR-145在宫颈鳞癌III~IV期与I~II期以及在宫颈鳞癌高、中分化与低分化组的表达均无明显差异(P>0.05).结论 miR-145在宫颈鳞癌组织中存在异常表达,提示其与宫颈鳞癌的发生及进展可能有关,值得进一步研究.%Objective To explore the expression of miR-145 in cervical intraepithelial neoplasia Ⅲ level ( CIN Ⅲ level) and cervical squamous carcinoma patients tissue and cervical squamous carcinoma , and to explore the relationship between the occurrence. Methods Real time RT-PCR method was used to assess the abundance of miR -145 in 49 cases of the cervical cancer tissues , CINⅢ levels tissues (includes CINⅢ levels 27cases and carcinoma 2 cases) and normal tissues (11 resectable cases) , which collected from Inner Mongolia Medical University Affiliated First Hospital . Results (1) The expression of the miR-145 was lower in the tissues of cervical cancer compared with that in the CINⅢ levels diseases and normal control ( P <0. 05 ). (2) No significant difference of miR-145 level was observed between different clinical stages ( stage Ⅰ~ Ⅱ and stages Ⅲ ~ Ⅳ) , different pathological type stage G1, stage G2 and stage G3 ( P >0. 05). Conclusion The miR445 in cervical squamous carcinoma tissue existed abnormal expression , suggested that it' s related with cervical squamous carcino -ma of the occurrence and progress , deserves further research.
