急性脑出血患者血肿周围组织IL-17和NF-κBp65的表达及意义

摘要

目的 探讨急性脑出血患者血肿周围组织白介素-17(interleukin-17,IL-17)和核因子-κBp65(nuclear factor-κBp65,NF-κBp65)的表达及意义.方法 选取2015年1月至2015年12月本院收治的急性脑出血患者血肿周围组织标本60例进行观察,根据患者发病至手术时间不同分为<6小时组、≥6小时且≤24小时组、>24小时且≤72小时组、>72小时组.选取本院同期脑出血远离血肿组织标本15例纳入对照组,并分别采用免疫组织化学染色、蛋白质印迹法及聚合酶链反应检测并比较不同组别患者血肿周围组织IL-17和NF-κBp65表达水平的差异.结果 急性脑出血患者标本中IL-17和NF-κBp65的表达水平较高,且随着出血时间的延长,IL-17和NF-κBp65阳性细胞占比先增后减;≤6小时组IL-17 Mrna表达水平与对照组比较无显著差异(P>0.05),≥6小时且≤24小时组、>24小时且≤72小时组、>72小时组IL-17 Mrna表达水平均显著高于对照组(P<0.01),其中>24小时且≤72小时组患者IL-17 Mrna表达水平最高.<6小时组、≥6小时且≤24小时组、>24小时且≤72小时组、>72小时组血肿周围组织中NF-κBp65表达水平均显著高于对照组(P<0.01),其中>24小时且≤72小时组NF-κBp65表达水平最高.结论 急性脑出血患者发病后血肿周围组织中IL-17和NF-κBp65的表达显著升高,以发病至手术时间>24小时且≤72小时最为明显.%Objective To investigate the expression and significance of interleukin-17 (IL-17) and nuclear factor-κBp65 (NF-κBp65) in the tissues around hematoma in patients with acute intracerebral hemorrhage. Method 60 samples from the tissues around hematoma with acute intracerebral hemorrhage in our hospital from January 2015 to December 2015 were selected, according to the different onset to peration time, they were divided into<6 hours group,≥6 hours and≤24 hours group,>24 hours and≤72 hours group, and>72 hours group. Another 15 samples away from the hematoma were selected as control group. The expression levels of IL-17 and NF-κBp65 were detected by immunohistochemistry, polymerase chain reaction and Western blotting.Result The expression levels of IL-17 and NF-κBp65 were higher in patients with acute cerebral hemorrhage. And the positive rate of IL-17 and NF-κBp65 cells increasedfirst and then decreased with the prolongation of bleeding time. The expression level of IL-17 between<6 hours group and control group had no significant difference. However, the expressions of IL-17 mRNA in the other groupswere higher than control group (P<0.01). Especially, the expression of IL-17 mRNA in>24 hours group and≤72 hours group was the highest. The expression levels of NF-κBp65 in≥6 hours and≤24 hours group,>24 hours and≤72 hours group,>72 hours group were significantly higher than that of control group (P<0.01). Especially, the expression level of NF-κBp65 in>24 hours and≤72 hours group was thehighest.Conclusion The expression level of IL-17 and NF-κBp65 in the tissues around hematoma with acute intracerebral hemorrhage significantly increased. The onset to operation time of>24 hours and≤72 hours is the most obvious.
