昆明山海棠对胶原性关节炎大鼠HIF-1α表达的影响及其作用机制

         

摘要

Objective To investigate the effect of tripterygium hypoglaucum hutch(THH) on expressions of hypoxia inducible factor (HIF) -1α in rats with collagen Ⅱ -induced arthritis(CIA) ,and its mechanism. Methods The established CIA rat model was given with THH for 30 d to observe the pathological changes of the joints with HE staining, detect HIF-1α expression by RT-PCR and immunohistochemical. Results 30 d later, HE staining showed the significantly reduced synovial proliferation dyeing and inflammation, immunohistochemical and PT-PCR displayed the obviously reduced HIF-1 α expression in THH group ( P < 0. 05 ). Conclusions THH has remarkably curative effect on CIA rats,which is relevant with the regulation of HIF-1α expression in peripheral blood and synovial organization.%目的 探讨昆明山海棠(THH)对低氧诱导因子(HIF-1α)在胶原性关节炎(CIA)大鼠模型中表达的影响及其作用机制.方法 建立CIA大鼠模型,然后用THH灌胃给药治疗30 d,观察关节炎指数(AI),30 d后HE染色观察关节的病理改变,RT-PCR以及免疫组化法检测HIF-1α的表达.结果 给药30 d后,THH组HE染色显示滑膜增殖以及炎症反应明显减轻,免疫组化以及PT-PCR显示HIF-1α表达明显降低(P<0.05).结论 THH对CIA大鼠有显著疗效,其作用机制与调节外周血以及滑膜组织内的HIF-1α的表达有关.
