茵陈蒿汤化裁方对高脂饮食小鼠肝组织肝脂酶活性及mRNA转录的影响

         

摘要

目的 观察茵陈蒿汤化裁方对高脂饮食小鼠血脂调节作用及肝组织肝脂酶(HL)活性及其mRNA转录水平的影响.方法 高脂饮食饲养小鼠6 w,并设普食对照、模型对照、茵陈蒿汤化裁方高、低剂量组.药物干预4 w后,生化检测血清总胆固醇(TC)、甘油三酯(TG)含量,张蓉法测定肝组织匀浆HL活性,RT-PCR法检测HL mRNA转录水平.结果 茵陈蒿汤化裁方可显著降低血清TG水平(P<0.01),提高肝组织匀浆HL活性(P>0.01),而对肝组织HL mRNA的转录水平无明显影响(P>0.05).结论 增强肝组织HL活性是茵陈蒿汤化裁方调节脂质代谢的药理学机制之一,该方在高脂血症、脂肪肝及动脉粥样硬化等疾病的中医药防治领域具有潜在的应用价值.%Objective To investigate the effects of fixed Yin-Chen-Hao decoction (FYCHD) on regulating the blood lipid and the activity and mRNA transcription level of hepatic lipase (HL) in liver tissues of mouse with high fat diet. Methods Mice were fed with high fat diet for 6 weeks and were randomly divided into model, FYCHD high and low dose groups. In addition, a routine diet was fed to mice in control group. All groups were treated with corresponding decoction or distilled water for 4 weeks. Levels of serum total cholesterol(TC) and triglyceride (TG) were determined. Activity and RT-PCR was adopted to measure the mRNA transcription level of HL in liver tissues. Results FYCHD could decrease remarkably the levels of serum TG(P <0. 01 ) and increase the activity of HL in liver tissues (P < 0. 01 ).But FYCHD couldn't change the mRNA transcription level of HL in liver tissues ( P > 0. 05 ). Conclusions Increasing the activity of HL in liver tissues is one of the mechanisms of FYCHD adjusting the lipid metabolism and FYCHD has potential value for the treatment of hyperlipidemia, fatty liver and atherosclerosis.
