缺氧条件下前列腺上皮细胞体外培养雌雄激素受体的表达

         

摘要

目的 观察在缺氧条件下前列腺上皮细胞体外培养细胞核膜表面雌雄激素受体表达的变化.方法 前列腺上皮细胞株RWPE-1体外培养,在缺氧或有氧培养环境中分别于接种后4、8、12、24、48 h检测其核膜雌雄激素受体基因和蛋白表达.基因表达采用RT-PCR法检测,蛋白表达采用免疫组化法检测.结果 缺氧条件下接种4h前列腺上皮细胞雄激素受体mRNA表达为(0.32±0.01),后随时间逐渐上升至48 h为(1.13±0.08),显著高于同期有氧条件下的(0.26±0.02,P=0.01)和(0.31±0.01,P=0.003);免疫组化显示,缺氧条件下雄激素受体蛋白表达自12h开始较有氧条件下显著增加,分别为(42.08±1.58)/200个细胞和(26.37±6.76)/200个细胞(P=0.017).有氧条件下前列腺上皮细胞不论在基因还是蛋白水平雌激素受体几乎不表达;而缺氧条件下12 h可检测出雌激素mRNA表达为(0.15±0.01),至48 h升至(0.41±0.04),4h时雌激素阳性表达上皮细胞为(4.98±0.49 )/200个细胞,后逐渐升至48 h的(45.94±0.90 )/200个细胞.结论 缺氧可刺激前列腺上皮细胞核膜表面雌雄激素受体表达的上调.%Objective To observe the different expression of androgen/eslrogen receptor in nucleus membrane of prostate epithelial cell at anoxic or normoxic conditions. Methods Prostate epithelial cell line RWPE-I was cultured in vitro. Al 4,8,12,24, and 48 h after the cell had been seeded,the gene and protein expressions of androgen receptor (AR)and estrogen receptor (ER) in prostate epithelial cell were tested by RT-PCR and immunohistochemistry. Results Significantly increasing expression of AR and ER could be seen in anoxic prostate epithelial cell compared with normoxic prostate epithelial cell. AR mRNA was (0.32±0.01) in anoxic prostate epithelial cell tested by RT-PCR at 4 h.and later increased to (1.13±0.08) at 48 h,which was higher than that of normoxic prostate epithelial cell (0.26±0.02) and (0.31±0.01) synchronously(P=0.0l and 0.003,respectively). There were(42.08±1.58) positive AR expression cells per 200 cells in anoxic condition at 12 h by immunohistochemislry,began significantly higher than (26.37±6.76) in normoxic condition (P=0.0l7)from 12 h. ER mRNA was (0.15±0.01) in anoxic prostate epithelial cell at 12 h.and increased to (0.41±0.04 ) at 48 h. There were (4.98±0.49) postive ER expression cells per 200 cells in anoxic condition at 4 h and increased to (45.94±0.90) at 48 h. But in normoxic condition,there were no gene and protein expression in prostate epithelial cell. Conclusions Anoxic prostate epithelial cell can upregulate the expression of androgen/estrogen receptor.
