小柴胡汤对大鼠肝细胞脂肪变性及葡萄糖调节蛋白表达的影响

         

摘要

目的 探讨小柴胡汤防治大鼠肝细胞脂肪变性的疗效及作用机制.方法健康Wister大鼠分为正常组、模型组及小柴胡汤低、高剂量组和东宝肝泰组,除正常组以外,其他各组采用四氯化碳联合高脂饮食诱导大鼠脂肪肝模型,各组大鼠给药干预4周后,采用HE染色观察肝组织病理变化,采用全自动生化仪及分光光度计测定血清中天冬氨酸氨基转移酶(AST)、丙氨酸氨基转移酶(ALT)及肝组织中胆固醇(TC)、三酰甘油(TG)的含量,采用RT-PCR和Western blot检测肝组织中葡萄糖调节蛋白78(GRP78)的表达.结果 小柴胡汤低、高剂量组的肝指数、血清ALT、AST水平及肝组织中TC、TG含量较模型组明显降低(P<0.05);病理HE染色显示,小柴胡汤组肝脂肪变性程度较模型组明显减轻;RT-PCR 和Western blot检测结果表明,小柴胡汤组肝组织中GRP78的表达显著低于模型组.结论 小柴胡汤具有抑制大鼠肝细胞脂肪变性的作用,其作用机制可能与内质网应激有关.%Objective To investigate the experimental efficiency and potential mechanisms of Xiaochaihu decoction in preventing fatty degeneration of liver cells in rats. Methods Healthy Wister rats were randomly divided into 5 groups: normal group, model group, Xiaochaihu decoction low and high dose group, and Dongbaogantai group. The model of fatty degeneration of liver cells in rats was established by carbon tetrachloride (CCU) subcutaneous injection along with high-fat diet in all groups except the normal group. After treatment for 4 weeks, HE staining was applied to detecting pathological changes of liver. The levels of serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), and total cholesterol (TC), triglyceride (TG) of liver were assayed by automatic biochemistry instrument and spectrophotometer. The expression of GRP78 in liver tissues was detected by RT-PCR and Western blot. Results The liver index, levels of serum ALT and AST, and the levels of TC and TG in Xiaochaihu decoction low and high dose groups were significantly decreased than those in model group (P<0.05). Pathological HE staining suggested that fatty degeneration of liver cells was notably slighter in Xiaochaihu decoction groups than in model group. The expression of GRP78 showed by RT-PCR and Western blot was reduced in Xiaochaihu decoction groups than in model group. Conclusion Xiaochaihu decoction can mitigate the fatty degeneration of liver cells in rats, which may be associated with endoplasmic reticulum stress.
