乙肝免疫球蛋白不能阻断乙肝病毒感染人绒毛膜癌细胞

         

摘要

AIM:To study the blocking effects of hepatitis B immunoglobulin( HBIG ) on hepatitis B virus ( HBV ) infection of choriocarcinoma JAR cells in vitro. METHODS:The JAR cells were divided into experimental groups, blank control group ( using fetal bovine serum ) and negative control group ( using healthy human serum). The experimental groups were divided into high concentration groups of HBIG ( HBV + 10 IU/L HBIG, or HBV + 10 IU/L HBIG ), minimum protective concentration group of HBIG ( HBV + 10 IU/L HBIG ), low concentration groups of HBIG ( HBV + 1 IU/L HBIG, or HBV +0. 1 IU/L HBIG ) and HBV group. The effect of HBIG on the cell growth was analyzed by MTT assay. HBV - DNA in JAR cells was determined by real - time fluorescence quantitative PCR. The expression of hepatitis B surface antigen( HBsAg) in JAR cells was detected by immunofluo re scent staining. RESULTS: Different concentrations of HBIG had no effect on the growth of JAR cells. No different of HBV - DNA copies in JAR cells infected with HBV and treated with different concentrations of HBIG was observed ( P > 0. 05 ). No difference of fluorescent intensity of HBsAg in JAR cells with HBV infection and treated with different concentrations of HBIG was found ( P >0. 05 ). CONCLUSION: HBIG dose not block the HBV infection of JAR cells in vitro.%目的:观察不同浓度乙肝免疫球蛋白(HBIG)在体外对乙肝病毒(HBV)感染人绒毛膜癌细胞系JAR细胞的影响.方法:实验分为HBIG实验组、空白对照组(胎牛血清)、阴性对照组(健康人血清).HBIG实验组又分为高浓度HBIG组(HBV+103IU/L HBIG、HBV+102 IU/L HBIG)、最低保护浓度HBIG组(HBV+10 IU/L HBIG)、低浓度HBIG组(HBV+1 IU/L、HBV+0.1 IU/L)和HBV组;采用MTT法检测HBIG对细胞生长的影响;采用实时荧光定量PCR检测细胞内HBV-DNA的表达;采用细胞免疫荧光染色法检测细胞内乙肝表面抗原(HBsAg)的表达变化.结果:(1) 不同浓度的HBIG对JAR细胞生长无影响;(2) 不同浓度HBIG组的细胞内HBsAg的荧光强度无显著差异(P>0.05);(3)不同浓度HBIG组的细胞内HBV-DNA无显著差异(P> 0.05).结论:HBIG不能阻断HBV感染体外培养的JAR细胞.
