可注射式纳米羟基磷灰石复合支架与骨髓间充质干细胞修复兔桡骨骨缺损

         

摘要

BACKGROUND:Nano-hydroxyapatite/chitosan/calcium sulphate hemihydrate (nHA/CS/CSH) composite scaffold constructed in preliminary experiments has good physical and chemical properties. n OBJECTIVE:To explore the effects of injectable nHA/CS/CSH scaffold co-cultured with bone marrow mesenchymal stem cels (nHA/CS/CSH/BMSCs) for repairing rabbit’s radial bone defects. n METHODS:Bone defects with the length of 10 mm were created on the middle of the bilateral radial bones of 18 New Zealand white rabbits. The defects were filed with the nHA/CS/CSH/BMSCs composite as treatment group, while the others were filed with HA as controls. The ability of repairing bone defects was evaluated by gross, X-ray and histopathological observation at 4, 8, 12 weeks after implantation. n RESULTS AND CONCLUSION:At the end of 12 weeks after implantation, in the treatment group, bone defects were completely repaired, the radial bone showed normal morphology, the bone marrow cavity was recanalized a ittle from both ends toward the middle, images in the defect area had no difference from normal bone tissues, a large amount of new lamelar bone tissues were visible, the Harvard’s system formed, the original defects were filed with newborn lamelar bone tissues, bone tissues were interconnected, and the bone marrow cavities was interlinked; in the control group, bone defect was repaired partly, the bone mineral density was lower than that of the normal bone tissue, the bone marrow cavities were recanalized partialy, and a few of new lamelar bone tissues, accompanied with connective tissues growing into the defects. The nHA/CS/CSH/BMSCs is effective for the repair of bone defects.%背景:前期实验构建的可注射性纳米羟基磷灰石/壳聚糖/半水硫酸钙三维复合支架材料具有良好的理化性质。n  目的:评估可注射性纳米羟基磷灰石/壳聚糖/半水硫酸钙复合骨髓间充质干细胞修复兔桡骨骨缺损的效果。n  方法:在18只新西兰白兔双侧桡骨中段建立长10 mm的节段性缺损,将注射性纳米羟基磷灰石/壳聚糖/半水硫酸钙/骨髓间充质干细胞复合材料植入一侧骨缺损,作为实验组;另一侧植入单纯羟基磷灰石材料,作为对照组,于植入第4,8,12周末分别行大体、X射线检查与组织学观察。n  结果与结论:植入第12周末,实验组骨缺损完全修复,桡骨形状基本恢复到术前形态,可见由两断端向中间有少许的骨髓腔再通,缺损区影像与正常骨组织已无区别,可见大量新生板层骨组织,哈佛氏系统形成,原缺损区被新生板层骨组织填充,骨组织相互连续,骨髓腔相通;对照组骨缺损未完全愈合,有部分骨皮质缺损,骨密度影略低于正常骨组织,部分骨髓腔再通,可见少量板层样骨组织形成,纤维组织填充。结果表明可注射式纳米羟基磷灰石/壳聚糖/半水硫酸钙人工骨复合骨髓间充质干细胞可促进骨缺损的修复。
