接枝槲皮素交联壳聚糖吸附树脂的合成及应用

         

摘要

Crosslinked chitosan-queroxy resin(QCCR) was synthesized by CCM and queroxy as substrates and 1,6-hexanediol diglycidyl ether (1,6-HDE) as the crosslinking agent. In this paper, the influence of temperature,time,pH and 1,6-HDE on the grafting ratio of queroxy was studied. QCCR was characterized by IR and TG-DTA. The equilibrium adsorption capacity of queroxy by QCCR is 61.30 mg/g under the initial concentration of 1.5 mg/mL, and the hydrophobic interaction was also characterized by AFM observation. Queroxy adsorbed could be eluted from QCCR by 95 % ethanol solution with pH 7 and elution flow velocity 1.5 mL/mim. And accordingly, elution rates reaches to 86.28%. Flavonoids content in buckwheat was up to 69.76%by this resin.%以戊二醛交联的壳聚糖微球(CCM)为载体、1,6-己二醇二缩水甘油醚(1,6-HDE)为交联剂,制备以槲皮素为吸附官能团的接枝槲皮素交联壳聚糖树脂(QCCR),研究了时间、温度及pH对接枝率的影响。通过红外光谱(IR),热重-差热分析(TG-DTA)进行了接枝有效性表征。QCCR在槲皮素浓度1.5 mg/mL的溶液中,对槲皮素的柱式吸附量达到61.30 mg/g,通过原子力显微镜(AFM)观察到黄酮母核之间的疏水相互作用。以pH=7、95%乙醇溶液以1.5 mL/min的流速洗脱吸附的槲皮素,洗脱率达到86.28%。以该树脂从荞麦提取液中吸附总黄酮,纯度达到69.76%。
