大蒜素在鲤、鲫血浆中的药物代谢动力学研究

         

摘要

This study was to establish a method for the determination of the garlicin by gas chromatography (GC) and in-vestigate the pharmacokinetic of garlicin in Cyprinus carpio specularis and Carassius auratus to provide theoretical basis for the reasonable medication scheme .After oral administration of garlicin with a dosage of 7.5 mg/kg body weight, plasma samples were taken at different time points and determined by gas chromatography (GC).The pharmacokinetic parameters were calculated by DAS3.0 software.The lowest detection limit of DADS and DATS was 10 ng/mL.The recovery rates were 77.05% ~97.98%, 73.39% ~108.51% in Carp tissue, and 87.05% ~95.68%, 83.69% ~102.75% in cru-cian carp respectively.Two main components of garlicin were good -absorbed and wide spread.DADS was absorbed more rapidly and eliminated more slowly than DATS .%建立鲤(Cyprinus carpio specularis)、鲫(Carassius auratus)血浆中大蒜素含量的气相色谱检测方法,并研究大蒜素在鲤、鲫体内的药物代谢动力学特征。对鲤、鲫以7.5 mg/kg 的剂量单次口灌大蒜素,于停药后0.25、0.5、0.75、1、1.5、2、4、6、8、12、24、36、48、72 h 采集血浆,采用气相色谱法(GC)检测药物含量,通过DAS3.0软件分析药动学参数。结果显示:二烯丙基二硫醚(DADS)和二烯丙基三硫醚(DATS)的最低检测限均为10 ng/mL , DADS、 DATS 在鲤血浆中的平均回收率分别为77.05%~97.98%、73.39%~108.51%,在鲫血浆中的平均回收率分别为87.05%~95.68%、83.69%~102.75%。药物的两种主要成分在健康鲤鲫体内吸收较完全,分布广泛, DADS 较 DATS 吸收快,消除慢。
