不同诱导子对油樟内生菌次生代谢产物的影响

         

摘要

[目的]提高油樟内生菌次生代谢产物的积累.[方法] 将不同诱导子(油樟油水浊液、油樟叶水提液和油樟愈伤组织)添加到油樟内生菌(YY26、YG42)的培养体系中,研究其对内生菌生长及其次生代谢产物(1,8-桉叶油素、松油烯-4-醇、α-松油醇)的影响.[结果]从内生菌生长情况来看,油樟愈伤组织对内生菌YY26的生长有明显抑制作用,而对YG42的生长有促进作用;油樟油水浊液对2种内生菌的生长都有一定的抑制作用;油樟叶水提液对2种内生菌的生长均无显著影响.从次生代谢产物积累来看,油樟叶水提液对2种内生菌产1,8-桉叶油素、松油烯-4-醇及α-松油醇具有较强的促进作用,且对1,8-桉叶油素的促进作用最强.当油樟叶水提液浓度为1.00%时,内生菌YY26的1,8-桉叶油素产量最高,比对照组提高了11.80倍;当油樟叶水提液浓度为2.50%时,内生菌YG42中的1,8-桉叶油素产量达到最大值,与对照组相比增加了7.73倍.油樟愈伤组织对总挥发性物质的促进作用最强,当其浓度为10.00%时,对内生菌YY26总挥发性物质的促进作用最强,比对照组提高了13.26倍.[结论]该研究在一定程度上提高了油樟内生菌(YY26、YG42)次生代谢产物的积累量,为其今后的工业化生产奠定了基础.%[Objective]To improve the accumulation of secondary metabolites of endophytic fungi of Cinnamomum longepaniculatum.[Method]Different inducers(oil, water extract and callus from C.longepaniculatum,were added in the culture system of endophytic fungi(YY26,YG42)of C.longepaniculatum to study their effects on the growth of endophytic fungi and their secondary metabolites(1,8-eucalyptol, terpinen-4-ol, α-terpineol).[Result]As for the growth of endophytic fungi, callus from C.longepaniculatum had obvious inhibitory effects on the growth of YY26,but it had promoting effects on the growth of YG42.Oil from C.longepaniculatum had some inhibiting effects on the growth of YY26 and YG42.Water extracts from C.longepaniculatum had no significant effect on the growth of YY26 and YG42.As for the accumulation of secondary metabolites,water extract from C.longepaniculatum had stronger promoting effects on the yield of 1,8-cineole, terpinen-4-ol and α-terpineol of YY26 and YG42, and it had the strongest promoting effects on the yield of 1,8-cineole.When the concentration of water extract was 1.00%,the yield of 1,8-cineole of YY26 was the highest, which was increased than that of control group by 11.80 times.When the concentration of water extract was 2.50%, the yield of 1,8-cineole of YG42 was the highest, which was increased than that of control group by 7.73 times.Callus from C.longepaniculatum had the strongest promoting effects on total volatile of secondary metabolites.When the concentration of callus from C.longepaniculatum was 10.00%,it had the strongest promoting effects on total volatile substances of YY26, which was increased than that of control group by 13.26 times.[Conclusion]The accumulation amount of secondary metabolites of YY26 and YG42 in C.longepaniculatum were improved, which can lay a foundation for its industrialized production in the future.
