香水百合再生体系的建立

         

摘要

[Objective ] To study the effects of different concentrations of exogenous hormones on the differentiation and multiplication of Lilium. Spp scale leaf and the forming of adventitious roots, so as to lay technical foundation for establishing its regeneration systerm. F Method] With Liliun. Spp scale leaf as the explant, the effects of different plant growth regulator combinations on the callus forming, the differentiation and multiplication of adventitious buds, and the forming of adventitious roots were discussed, and the transplanting technique was studied. [Result] In the medium of MS +0. 5 - 1.0 mg/L 6-BA +0.1 -0. 3 mg/L IAA, the effect on differentiation and multiplication of sprouts was good; the differentiation rate reach above 78. 5% and multiplication rate reached over 5.6; inl/2MS+0.1 mg/L6-BA+0.5 mg/L IAA or 1/2MS +0. 1 mg/L 6-BA +0.1 -0.3 mg/L NAA or 1/2MS +0. 1 mg/L 6-BA +0. 8 mg/L IBA, the rooting result was good and the rooting rate could be up to96.0% ; using the substrate of 1:1 peat: perlite, the survival rate could reach a level of 96. 3 % . [ Conclusion ] The study on the isolated culture of Lilium. Spp had great significance to the rapid and scale seedling culture and realizing its genetic transformation by genetic engineering technique.%[目的]探讨不同外源激素水平对香水百合小鳞茎分化、增值及不定根形成的影响,为其再生体系的建立奠定技术基础.[方法]取香水百合鳞叶为外植体,探讨不同植物生长调节剂组合对其离体培养中愈伤形成、不定芽分化、增值、不定根形成的影响,并对试管苗移栽技术进行研究.[结果]香水百合小鳞茎分化与增值培养以MS+0.5~1.0mg/L6-BA+0.1~0.3 mg/LIAA效果好,分化率达78.5%以上,增值率可达5.6以上;生根以1/2 MS +0.1 mg/L 6-BA +0.5 mg/L IAA或1/2 MS +0.1 mg/L 6-BA +0.1~ 0.3 mg/L NAA或1/2 MS +0.1 mg/L 6-BA +0.8 mg/L IBA效果好,生根率可达96.0%;在泥炭:珍珠岩=1:1的基质中驯苗,成活率可达96.3%.[结论]香水百合离体培养技术的研究对其规模化快速育苗及进一步利用基因工程技术实现香水百合遗传转化具有重要意义.
