S-8大孔吸附树脂分离纯化洒金桃叶珊瑚中桃叶珊瑚苷的工艺研究

         

摘要

[Objective] To study the separation and purification process of aucubin with macroporous resin, so as to lay foundation for preparing aucubin from Aucuba japonica Variegata. [ Method] Taking aueubin as index, the concentration of aucubin was measured by HPLC to optimize the best separation and purification conditions of aucubin with macroporous resin. [ Result] The optimum conditions of S-8 macroporous resin were as follows; sample concentration of 2.76 mg/ml, sample flow velocity of 2 BV/h, sample capacity of 9.20 mg/g, elutant of 15% ethanol aqueous, elution flow rate of 2 BV/h, and total quantity of eluant of 3 BV. [ Conclusion] It has good effect to separate aucubin by S-8 macroporous resin, the content and recovery rate of aucubin were 48. 04% and 82. 30% .%[目的]研究大孔吸附树脂分离纯化桃叶珊瑚苷的工艺,为进一步精制洒金桃叶珊瑚中桃叶珊瑚苷奠定基础.[方法]以桃叶珊瑚苷为指标成分,用HPLC法测定桃叶珊瑚苷的含量,并优选大孔吸附树脂分离纯化桃叶珊瑚苷的最优工艺.[结果]S -8大孔吸附树脂分离纯化桃叶珊瑚苷的最优条件为:上样液浓度2.76 mg/ml,上样液流速2 BV/h,上样量9.20 mg/g,洗脱溶媒为浓度15%乙醇,洗脱流速2 BV/h,洗脱剂用量3 BV.[结论]S-8大孔吸附树脂对桃叶珊瑚苷分离纯化效果良好,产品纯度为48.04%,回收率为82.30%.
