[ Objective ] To establish a new rubber tree genetic transformation system for the use of gene engineering technology to research rubber tree. [Method] The leaves and stems of rubber tree Reyan 7-33-97 were infected by Agrobacterium rhizogenes R1601, 15834 and 1. 2556 to induce the hairy roots, and then the PCR technique was used to detect the hairy roots induced by Agrobacterium rhizogenes. [ Result] Under the same conditions, the hairy roots can be only successfully induced from rubber tree stems by Agrobacterium rhizogenes R1601, and its induction rate reached 36. 6% ; PCR results showed that T-DNA sequences have been integrated into the genome of rubber tree. [ Conclusion] An Agrobacterium rhizogenes mediated rubber tree genetic transformation system has been preliminarily established.%[目的]建立新的橡胶树遗传转化体系,为利用基因工程技术研究橡胶树提供基础.[方法]利用发根农杆菌R1601、15834和1.2556 3个株系分别侵染橡胶树热研7- 33 - 97的叶片和茎段,诱导毛状根,并利用PCR技术检测诱导得到的毛状根.[结果]在相同的条件下,仅橡胶树茎段在发根农杆菌R1601侵染后产生毛状根,其诱导率达36.6%;PCR结果表明,T-DNA序列已整合进橡胶树的基因组中.[结论]初步建立了发根农杆菌介导的橡胶树遗传转化体系.
