The bone powder is produced with the jointly use of low temperature freezing and biological enzymatic hydrolysis. On the basis of single factor test, response surface method which is based on the central composite experimental design principle is used to optimize the process for the preparation of bone powder with the weighted average (comprehensive score) of the bone yield and bioavailability as index. Optimal preparation process conditions are enzyme dosage 0.22 g, time of enzymatic hydrolysis 4.20 h, temperature of enzymatic hydrolysis 41℃, at which the comprehensive score is 76.23.%以牛骨为原料,采用低温冷冻和生物酶解相结合的技术制备超微骨粉。在单因素实验基础上,根据中心组合实验设计原理采用响应面实验法,以超微牛骨粉得率和钙生物利用率算术加权(即综合得分)为考察指标,对制备的技术参数进行优化。研究结果表明:实验范围内,最佳的技术参数为酶用量0.22 g,酶解时间4.20 h,酶解温度41℃;在该条件下,综合得分最高为76.23。
