大鼠精索静脉曲张对睾丸生精细胞Cx43表达的影响

     

摘要

Objective To study the effect of experimental varicocele on the Cx43 expression in adolescent rats’ testis spermatogenic cells. Method A rat varicocele model was built by partially ligating the rat ’ s left renal vein. 30 Male with SD rats were randomly divided into 2 groups, control group and varicocele group ( surgery group) ,15 rats in each group,and then each group was further divided into three sub-groups,5 rats in each sub-group;the ligation lasted for 2 weeks,4 weeks and 8 weeks,respectively;at the corresponding time points,the left testis of rats in each group were removed, then RT-PCR was applied to detect the Cx43 and HIF-1α gene expressions,and on week 8, the Cx43 and bcl-2 protein levels were detected by Western-blot assay Cx43. Results In the control group, there was no significant change in Cx43 and HIF-1α gene expression at the different time points,and in the surgical group,the HIF-1α gene expression was increased and the Cx43 gene expression was decreased, showing statistically significant differences compared with those in the control group ( P<0 . 05 );protein assay results showed that Cx43 and bcl-2 protein expressions were significantly decreased on week 8 in the surgical group. Conclusion Varicocele can cause the hypoxia in rat testis,the expression of Cx43 and bcl-2 down-regulate, and induce the apoptosis of the testis spermatogenic cells, thereby affecting the spermatogenesis of rats’ testes.%目的:探讨精索静脉曲张( VC)对青春期大鼠睾丸生精细胞Cx43的影响.方法部分结扎大鼠左肾静脉,建立精索静脉曲张模型.取雄性SD大鼠30只,随机分为2组,每组15只,分为对照组、精索静脉曲张组(手术组),每组中再分为3个亚组,每组5只,结扎持续时间为2,4,8周;达到相应的时间段后取各组大鼠左侧的睾丸,运用RT-PCR法检测Cx43及HIF-1α基因的表达,并在8周时通过Western-blot法检测Cx43及bcl-2蛋白表达水平.结果对照组在2,4,6周时Cx43及HIF-1α基因表达无明显变化;手术组在2,4,6周时HIF-1α基因表达水平提高,Cx43基因表达水平下降,与对照组相比,差异具有统计学意义(P<0.05);蛋白检测结果表明:8周时手术组Cx43及bcl-2蛋白表达水平显著下降.结论精索静脉曲张可致大鼠睾丸组织缺氧,Cx43及bcl-2表达下调,并诱导睾丸组织生精细胞凋亡增加,进而影响睾丸生精功能.
