高表达单克隆抗体ER的细胞株筛选

         

摘要

ER is monoclonal antibody of type of IgGl, which is monoclonal chimeric antibody come from human and mouse.ER is on inhibitor to epidermal growth factor receptor. Investigations were made to study the process optimiza-tion of CHO cell culture, including cell weaning , screening cell lines which have high expression of the monoclonal antibody ER and using protein—free medium to improve cell concentration and MAb production. The viable cell concen-tration, and the MAb production reached 6.8*106 cells/ml and 117 mg/1, respectively%ER为IgG1单克隆抗体,是一种人和鼠的EGFR单克隆抗体的嵌合体.ER是表皮生长因子受体(EGFR)阻止剂,可以竞争性的抑制EGF及其它配体的结合.在本课题研究中以CHO细胞为对象,进行细胞驯化,无蛋白培养基的培养,筛选高表达细胞株,旨在通过上述条件优化,提高细胞密度和单抗产量.实验结果为最高活细胞密度以及单抗产量分别达到了6.8*106cells/ml和117mg/l.
