山楂叶及其提取物中2″-O-鼠李糖牡荆素和牡荆素的含量测定

         

摘要

目的建立对山楂叶及其提取物中有效成分2″-O-鼠李糖牡荆素和牡荆素的含量同时进行测定的方法.方法应用反相高效液相色谱法同时测定山楂叶及其提取物中2″-O-鼠李糖牡荆素和牡荆素的含量.以C18柱作为色谱柱,四氢呋喃-乙腈-水-磷酸(30∶5∶125∶0.1)为流动相,检测波长为270 nm,按外标法定量.结果线性范围分别为0.106 4 μg~2.128 0 μg(相关系数r =0.999 1)和0.139 2~2.784 0 μg (r = 0.999 3).在山楂叶药材中,2″-O-鼠李糖牡荆素和牡荆素的平均加样回收率为99.21%(RSD = 2.80%, n = 6)和100.56%(RSD = 2.84%, n = 6);重复性试验的RSD分别为2.74%(n = 6)和1.99%(n = 6).结论本方法简便,分离重现性好,灵敏度高,为山楂叶及其提取物中2″-O-鼠李糖牡荆素和牡荆素的含量同时测定提供了可行的含量测定方法,为山楂叶的质量标准提供了测定依据.%Aim To establish an RP-HPLC method for simultaneous determination of 2″-O-rhamnosyl vitexin and vitexin in Chinese hawthorn leaf and its extract. Methods Chromatography was carried out on Kromasil C18 column (250 mm × 4.6 mm, 5 μm), using THF-CH3CN-H2O-H3PO4 (30∶5∶125∶0.1) as the mobile phase at a flow rate of 1.0 mL·min-1. The UV detection wavelength was 270 nm. Results The linear range of 2″-O-rhamnosyl vitexin and vitexin were 0.106 4 μg -2.1280 μg ( r =0.999 1 ) and 0.139 2 μg - 2.784 0 μg ( r =0.999 3 ), respectively. The average recoveries of 2″-O-rhamnosyl vitexin and vitexin in Chinese hawthorn leaf were 99.2% (RSD = 2.80%, n= 6) and 100.6% (RSD = 2.84%, n = 6), respectively. Conclusion This method is reproducible, simple, precise, and rapid for simultaneous determination of 2″-O-rhamnosyl vitexin and vitexin in Chinese hawthorn leaf and its extract, thereby providinge the basis for quality specification of Chinese hawthorn leaf and its extract.
