The phospholipid was extracted from the egg of large yellow croaker (Pseudosciaena crocea) using hot ethanol. The effects of ethanol concentration, the ratio of material to liquid, extraction temperature, extraction time, extraction frequency on product yield were investigated. Through orthogonal design the optimum conditions were determined. The results showed that the optimal conditions were ethanol content 92% , the ratio of material to liquid 1:8, extraction temperature 45 ℃, extraction time 20 min, and the frequency was 3. Under the conditions, the product yield was 47.73%. Using thin layer chromatography combined with spectro-photometry to test the components of phospholipid, the phospholipid from the egg of large yellow croaker included phosphatidylcho-line, phosphatidylethanolamine, phosphatidylinoitide, springomyelin, lysophosphatide, the relative contents were 69.38%, 4.14%, 8.52% , 7.24% , 10.72% respectively.%采用乙醇加热纯化提取的方法,从大黄鱼鱼卵中提取磷脂,研究乙醇含量、料液比、浸提温度、浸提时间、浸提次数对磷脂提取率的影响,并用正交设计优化提取工艺条件.结果表明,最佳工艺条件为:乙醇含量为92%、料液比1∶8、浸提温度45℃、浸提时间20min、浸提3次,在该条件下产品的提取率可达47.73%.采用薄层层析法结合分光光度法分析测定大黄鱼鱼卵磷脂含磷脂酰胆碱、磷脂酰乙醇胺、磷脂酰肌醇、神经鞘磷脂、溶血磷脂酰胆碱5种组分,相对含量分别为69.38%、4.14%、8.52%、7.24%、10.72%.
