HPLC测定动物源性食品中倍他米松的残留

         

摘要

为建立检测动物性食品中倍他米松残留的高效液相色谱法。样品经乙酸乙酯提取后,进行液相色谱分析。采用Agilent C18(250 mm×4.6 mm,5μm)色谱柱,流动相为甲醇:水(60∶40,v∶v),流速为0.8 mL/min,检测波长为240 nm,柱温为30℃。方法的检出限和定量限分别为0.152μg/kg 和0.502μg/kg。倍他米松在0.5~32μg/mL范围内线性关系良好,相关系数为0.9989;选取5种动物源性食品进行倍他米松的加标回收试验,回收率在72.73%~93.06%之间,精密度RSD为3.78%(n=6)。本方法简便、可靠、重复性好,可用于动物性食品中倍他米松残留的检测。%The research aims to establish a high performance liquid chromatography(HPLC)method to detect betamethasone in animal tissues.The sample was extracted by ethyl acetate and analyzed by HPLC.An Agilent C18(250 mm×4.6 mm,5μm)cartridge was used as chromatographic column. The mobile phase was methanol∶water (60∶40 ),flow rate 0.8 mL/min,detection wavelength 240 nm and column temperature 30℃.The limit of detection and quantity were 0.152 μg/kg and 0.502μg/kg.The average recovery rate of betamethasone in 5 animal tissues were in the range of 72.73%~93.06%with the precision of 3.78%(n=6).The method is simple,reliable and reproduc-ible,and it is suitable for the determination of betamethasone residue in food of animal tissues.
