根据木聚糖酶与木聚糖酶抑制蛋白之间的特异性结合作用,采用固定化木聚糖酶在不同pH值、温度和时间下对小麦籽粒粗蛋白提取液中的木聚糖酶抑制蛋白进行亲和吸附,并通过改变pH值对木聚糖酶-木聚糖酶抑制蛋白复合物进行解吸研究,以期为木聚糖酶抑制蛋白的获得提供一个较好的新方法.结果表明,固定化木聚糖酶与木聚糖酶抑制蛋白的最佳吸附条件为:小麦籽粒粗蛋白提取液pH值6.5、反应温度30℃、反应时间50 min;固定化木聚糖酶-木聚耱酶抑制蛋白复合物的最佳解吸条件为:巴比妥钠-盐酸缓冲液pH值8.5.%Based on the special interaction between xylanase and xylanase inhibitor protein,inhibitor protein was purified from the protein crude extract of wheat seeds using affinity chromatography with immobilized xylanse under the conditions of different pH values,temperatures and times.Subsequently,the desorption experiment of enzyme-inhibitor complex was carried out under different pH values.The results showed that the optimum adsorption pH value was 6.5,temperature was 30 ℃,time was 50 min; the optimum desorption pH value was 8.5.Purification of inhibitor protein using affinity chromatography with immobilized xylanse is a better new method.
