枸杞多糖对胃癌模型大鼠免疫功能的影响

         

摘要

目的:探讨枸杞多糖对胃癌模型大鼠免疫功能的影响.方法:将50只SD大鼠随机分为正常对照组、胃癌模型组、枸杞多糖(LBP)低剂量组(5 mg·kg-·1d-1)、LBP中剂量组(10 mg·kg-·1d-1)、LBP高剂量组(20 mg·kg-·1d-1),每组各10只,经灌胃给药,1次/d,持续10 d.测定大鼠抑瘤率、胸腺指数、外周血CD4+T细胞比例、CD4+CD25+T细胞比例、DC致敏的细胞毒性T淋巴细胞(CTL)杀伤功能及外周血白介素2(IL-2)、肿瘤坏死因子α(TNF-α)水平.结果:与正常对照组比较,胃癌模型组、LBP治疗组胸腺指数、CD4+T细胞比例、CTL杀伤功能、IL-2水平下降,但不同浓度LBP组均高于胃癌模型组;胃癌模型组、LBP治疗组CD4+CD25+T细胞比例、TNF-α水平升高,但不同浓度LBP组低于胃癌模型组,差异有统计学意义.与LBP中剂量组比较,较LBP低、高剂量组抑瘤率、胸腺指数、CD4+T细胞下降、CTL杀伤功能、IL-2水平下降,CD4+CD25+T细胞比例、TNF-α水平升高,但LBP低、高剂量组间比较差异无统计学意义.结论:LBP可提高胃癌大鼠免疫功能,发挥抗肿瘤作用.%Objective To investigate the effect of lycium barbarum polysaccharide (LBP) on immune function in rats with gastric cancer. Methods 50 SD rats were randomly divided into the normal control group, gastric cancer model group, low dose LBP group (5 mg·kg-·1d-1), medium dose LBP group (10 mg·kg-·1d-1) and high dose LBP group (20 mg·kg-·1d-1), 10 rats in each group. All rats were administrated by gavage, once /d, for 10 days. The tumor inhibitory rate, thymus index, proportion of peripheral blood CD4+T cells, proportion of CD4+CD25+T cells, DC sensitized cytotoxic T-lymphocyte (CTL) killing function and levels of peripheral blood interleukin 2(IL-2) and tumor necrosis factor α (TNF- α) were determined. Results The thymus index, proportion of peripheral blood CD4+T cells, CTL killing function and IL-2 level were lower in the gastric cancer model group and LBP treatment group than the normal control group, and they were higher in different concentrations of LBP groups than gastric cancer model group. The proportion of CD4+CD25+T cells and TNF- α levels increased in the gastric cancer model group and LBP treatment groups, and those were lower in different concentrations of LBP groups than gastric cancer model group. Compared with medium dose LBP group, the tumor inhibitory rate, thymus index, proportion of CD4+T cells, CTL killing function and IL-2 levels were lower in the low dose and high dose LBP groups, while the proportion of CD4+CD25+T cells and TNF- α levels were higher in the low dose and high dose LBP groups. There was no statistically significant difference between the low dose group and high dose LBP group. Conclusion LBP can improve the immune function of rats with gastric cancer and play an anti-tumor role.
