蜘蛛丝蛋白促凝血作用及其机制的初步探讨

         

摘要

Objective To investigate the procoagulant role of spider silk protein and discuss its mechanism. Methods Tocompare the clotting time in vitro and hemostatic time of spider silk protein group, Yunnan Baiyao group and saline groupthrough coagulation experiment conducted in vitro (coagulation method, tube method and recalfication time) and hemostatic experimentin wound surfaces (skin, liver, and femoral artery) in SD rats. Activated partial thromboplastin in time (APTT), prothrombintime (PT) and thrombin time (TT) were evaluated by solidification method in rats of each group. Results The clottingtime in vitro and hemostatic time of spider silk protein group and Yunnan Baiyao group were obviously shortened comparedwith that of saline group (P<0.01 or P<0.05). Moreover, APTT, PT and TT were also obviously shortened compared with thoseof saline group (P<0.01 or P<0.05). Conclusion Spider silk protein shows obvious blood clotting effect. Activating endogenouscoagulation and extrinsic coagulation may be one of its procoagulant mechanisms.%目的 探索蜘蛛丝蛋白的促凝血作用,初步探讨其作用机制.方法 通过体外凝血实验(凝血板法、试管法、血浆复钙时间测定)及创面止血实验(以SD大鼠为研究对象,进行皮肤创面、肝脏创面、股动脉切开进行局部创面止血实验),比较蜘蛛丝蛋白组、 云南白药组、 生理盐水组各组的凝血时间及止血时间;并采用凝固法测定各组大鼠活化部分凝血活酶时间(APTT)、凝血酶原时间(PT)和凝血酶时间(TT).结果 与生理盐水组比较,蜘蛛丝蛋白组与云南白药组均能显著缩短大鼠体外凝血时间及大鼠皮肤创面、肝脏创面及股动脉切口的止血时间(P<0.01或P<0.05),且能缩短大鼠的APTT、PT和TT(P<0.01或P<0.05).结论 蜘蛛丝蛋白具有较明显的促凝血作用,激活内源性凝血系统和外源性凝血系统可能为其作用机制之一.
