1株产单宁酶细菌的分离鉴定及产酶条件研究

         

摘要

从富含单宁酸的土壤中分离筛选出1株产单宁酶的细菌,经过菌落形态观察和16S rDNA分子生物学鉴定,该细菌为肺炎克雷伯菌。对该菌所产胞外单宁酶的发酵条件进行了初步研究,得出最佳产酶条件:培养温度37℃,培养时间36 h,培养转速180 r/min,单宁酸含量2 g/L,最佳碳源为葡萄糖,最佳氮源为氯化铵,此时所产单宁酶活力为0.8 U/mL。%A tannase-producing strain isolated and screened from tannic-acid-rich soil, it was identified as Klebsiella pneumoniae based on the observation of its colony morphology and the molecular biological classification of its 16S rDNA. The fermentation conditions for tannase production of the strain were initially studied. The results showed that the optimal conditions were 37 ℃, cultivated for 36 h at 180 r/min;the content of tannic acid was at 2 g/L, the best carbon source was glucose, and the best nitrogen source was ammonium chloride. The produced tannase activity was at 0. 8 U/mL under the conditions.
