不同稀释液对检测血清乳酸脱氢酶的影响

         

摘要

Objective To investigate the influence of different dilution solutions on serum LDH detection by comparing the changes of serum LDH after specimen dilution. Methods The AU5800 automatic biochemical analyzer was used to detect the LDH concentrations before and after specimen dilution. The detection results of the same high LDH specimen were compared between the original concentration and concentrations after dilution by 3 kinds of dilution solutions of deionized water ,normal saline and inactivated serum. The degree of regression coefficient (RC) close to 1 was compared by drawing the regression curve. Results When 2 times of dilutions,RC of deionized water(RCbw)=1.038 4,RC of normal saline(RCbs)=1.043 4,RC of inactivated serum(RCbi)=1.023 4;when 5 times of dilutions,RCbw=1.152 9,RCbs=1.144 4,RCbi=1.142 0;when 10 times of dilutions,RCbw=1.107 8, RCbs=1.112 4,RCbi=0.995 8. Conclusion In the comparison of 2,5,10 of 3-dilution gradients,RCbi is most close to 1 compared with RCbs and RCbw,indicating that inactivated serum is the best dilution solution among these 3 kinds of dilution solution.%目的:通过比较标本稀释前后血清乳酸脱氢酶(LDH)的变化,探讨不同稀释液对检测血清LDH的影响。方法用AU5800全自动生化分析仪检测标本稀释前后的LDH水平,比较用去离子水、生理盐水和灭活血清3种稀释液稀释同一份高LDH标本后与原始浓度的检测结果,通过绘制回归曲线,比较回归系数接近1的程度。结果2倍稀释时:去离子水回归系数(b水)=1.0384,生理盐水回归系数(b盐)=1.0434,灭活血清回归系数(b灭)=1.0234;5倍稀释时:b水=1.1529,b盐=1.1444,b灭=1.1420;10倍稀释时:b水=1.1078,b盐=1.1124,b灭=0.9958。结论通过2、5、10倍3个稀释梯度的比较,b灭相对b盐、b水最为接近1,说明灭活血清是三者中最好的稀释液。
