Objective:To examine the effects of ellagic acid(EA)on estrogen - induced cell proliferation via at-tenuating extracellular - signal - regulated kinase(ERK)1 / 2 activity. Methods:The key factor and mechanism of the effects of EA on breast cancer were investigated by evaluating estrogen - induced cell proliferation by cell prolifer-ation assay and phosphorylation of ERK1 / 2 by Western blot. Results:Estrogen induces ERK1 / 2 activation on time dependent manner in MCF - 7 breast carcinoma cells. MCF - 7 cell proliferation was dramatically increased about 3 -fold when cells were exposed to estrogen. Estrogen - induced ERK1 / 2 activation was predominantly inhibited by EA on dose dependent manner. Inhibition of ERK1 / 2 activation by EA led to significant suppression of cell proliferation. Conclusion:Estrogen induces the proliferation of MCF - 7 cells partially via activation of ERK1 / 2 pathway,and this effect appears to be inhibited by EA.%目的:探讨鞣花酸在雌激素诱导的乳腺癌细胞增殖中的作用及细胞和分子机制。方法:雌激素培养乳腺癌细胞 MCF -7,鞣花酸培养液处理,观察细胞增殖情况。采用 Western blot 观察细胞外调节蛋白激酶1/2(ERK1/2)的表达。结果:雌激素可明显促进 MCF -7细胞增殖,与对照组比较有显著性差异,并能激活 ERK1/2,使 p - ERK1/2表达明显增加,且具有时间依从性。鞣花酸可明显抑制雌激素诱导的 MCF -7细胞增殖,并抑制 p - ERK1/2的表达,呈剂量依赖性。结论:雌激素通过激活 ERK1/2诱导 MCF -7细胞增殖,而这一效应可被鞣花酸抑制。
