NF-κB 介导热休克蛋白70减轻大鼠急性坏死性胰腺炎的实验研究

         

摘要

目的:探讨热休克蛋白70(heat shock protein 70,Hsp70)减轻大鼠急性坏死性胰腺炎的机制。方法将60只 SD 大鼠按随机数字表法分为3组,每组20只。A 组急性坏死性胰腺炎(aute nicrosis apoptonisis,ANP)模型由5%牛黄胆酸钠大鼠胰胆管逆行注射诱发而成,B 组手术进腹后翻动十二指肠,C 组使用温生理盐水持续腹腔灌洗30 min,10 h 后同 A 组造 ANP 模型。于最后1次手术后6 h 处死大鼠,检查各组大鼠血清细胞因子 IL-6、TNF-α和 IL-8及血清淀粉酶;取胰腺组织行病理检查,并分别使用 PCR 和免疫组织化学方法检测胰腺中 Hsp70和 NF-κB 的表达。结果C 组大鼠血清淀粉酶为(2845.70±204.78)U·L-1,明显低于 A 组的(4064.93±188.97)U· L-1,高于 B 组的(1221.47±54.58)U·L-1(均 P <0.05);C 组大鼠血清 TNF-α、IL-6、IL-8明显低于 A 组,高于B 组(均 P <0.05);C 组大鼠胰腺组织 Hsp70 mRNA RQ 值明显低于 A 组,但高于 B 组(P <0.05);NF-κB 在胰腺中的表达:A 组部分细胞核强阳性染色,胞浆中弱阳性染色,B、C 组细胞核染色明显减弱,胞浆则未见染色。结论HSP70可以通过调控 NF-κB 从而下调细胞因子的表达,进而改善大鼠 ANP 病理生理。%Objective To explore the mechanism of action of heat shock protein 70(Hsp70)in relieving acute necrotizing pancreatitis(ANP).Methods Sixty SD rats were randomly divided in-to three groups,with 20 rats in each group.In group A,ANP was induced by retrograde injection of 5% sodium taurocholate into pancreatic duct.In group B,the duodenum was flipped after ab-dominal surgery for the sham operation.In group C,ANP was induced 10 hours after continuous peritoneal lavage with warm saline for 30 minutes.The rats were sacrificed 6 hours after the last operation.Serum levels of tumor necrosis factor-α(TNF-α),interleukin-6(IL-6),interleukin-8(IL-8)and amylase were measured by ELISA.In addition,pancreas tissues were obtained to examine the pathological changes.HSP70 expression was examined by PCR and nuclear factor-κB(NF-κB) expression by immunohistochemistry.Results The levels of serum amylase in group C[(2 845.70± 204.78)U·L-1 )]were lower than those in group A[(4 064.93±188.97)U·L-1 ],but higher than those in group B[(1 221.47±54.58 )U·L-1 ](all P <0.05);The levels of serum TNF-α, IL-6 and IL-8 in group C were lower than those in group A,but higher than those in group B(all P <0.05);The RQ of Hsp70 mRNA in group C was lower than that in group A,but higher than that in group B(P < 0.05);Immunohistochemistry showed strong positive nuclear staining and weak positive cytoplasmic staining for NF-κB in group A,and weak positive nuclear staining and negative cytoplasmic staining for NF-κB in group B and C.Conclusion HSP70 can decrease cyto-kine expression and reduce pathophysiological damage through down-regulating the expression of NF-κB in ANP rats.
