SPHK1在舌鳞癌中的表达及其表达下调对Tca8113细胞迁移能力的影响

         

摘要

Objective: To investigate the expression of sphingosine kinase-1 (SPHK1) in tongue squamous cell carcinoma tissue, and to analyze the effects of SPHK1 downregulation on the migration of tongue squamous cell carcinoma Tca8113 cells. Methods: Expres-sion oi SPHK1 protein was detected by immunohistochemistry in 45 cases of tongue squamous cell carcinoma tissue, 36 cases of dyspla-sia tissue and 45 cases of normal tongue epithelial tissue. SPHK1 siRNA and control siRNA were transfected to tongue squamous cell carcinoma Tca8113 cells by Lipofectamine 2000 respectively. SPHK1 protein expression in the cells was investigated by Western blotting. Cell proliferation and migration were examined by CCK-8 kit and Boyden chamber respectively. MMP-2 and MMP-9 proteins were analyzed by Western blotting. Results: Expression of SPHK1 protein in tongue squamous cell carcinoma tissue was higher than that in dysplasia tissue and normal tongue epithelial tissue(χ2 =55.608, P =0.000). SPHK1 siRNA obviously downregulated the expression of SPHK1 protein in Tca8113 cells, the downregulation inhibited the proliferation and migration of Tca8113 cells. The down-regulation of SPHK1 expression decreased the expressions of MMP-2 and MMP-9 proteins. Conclusion; SPHK1 overexpression may play a major role in the occurrence and development of tongue squamous cell carcinoma. The migration decrease of Tca8113 cells mediated by the down-regulation of SPHK1 expression may be associated with the down-regulation of MMP-2 and MMP-9 proteins.%目的:研究鞘氨醇激酶1(SPHK1)在舌鳞癌组织中的表达及其表达下调对Tca8113细胞迁移的影响.方法:采用免疫组织化学检测45例舌鳞癌组织、36例癌旁异常增生组织和45例正常舌上皮组织中SPHK1蛋白的表达.利用脂质体2000将SPHK1 siRNA和对照siRNA分别转染Tca8113细胞,Western blotting检测细胞中SPHK1蛋白的表达;分别采用CCK-8和Boyden小室检测细胞增殖和细胞迁移能力的变化;用Western blotting检测细胞迁移相关蛋白MMP-2和MMP-9表达的变化.结果:舌鳞癌组织中SPHK1蛋白的表达显著高于癌旁异常增生组织和正常舌上皮组织(X2=55.608,P=0.000).SPHK1 siR-NA能显著下调Tca8113细胞中SPHK1蛋白的表达,其表达下调能明显抑制舌鳞癌细胞的增殖和降低细胞的迁移能力.SPHK1表达的下调能明显降低舌鳞癌细胞中MMP-2和MMP-9蛋白的表达.结论:SPHK1的过表达可能在舌鳞癌的发生发展过程中发挥重要作用,其表达下调介导的舌鳞癌细胞迁移能力的降低可能与MMP-2和MMP-9的下调密切相关.
