Objective: To investigate the effects of COX-2 selective inhibitor NS-398 on a human cervical carcinoma cell line, Hela, and explore the possibility of its application in anticancer treatment.Methods: Human cervical carcinoma Hela cells were treated with different concentrations of NS-398.The inhibitory effect of NS-398 on proliferation of Hela cells was assessed by Cell Counting Kit-8 (CCK-8) method.Rhodanmine 123 staining was applied to detect mitochondrial membrane potential changes in the cells, and agarose gel electrophoresis was used to show DNA ladders.Results: NS-398 significantly inhibited the growth of Hela cells.The inhibition rate increased as the concentration and treatment time increased.The mitochondrial membrane potential reduced significantly when Hela cells were treated with NS-398 for 24 hours and this effect was closely related to the concentration of NS-398.Apoptosis was observed in Hela cells treated with NS-398 for 48 hours.Conclusions: NS-398 significantly inhibited the growth of cervical carcinoma cells and may induce the apoptosis of the cells through a mitochondrial pathway.%目的 探讨环氧化酶-2(COX-2)选择性抑制剂NS-398诱导宫颈癌细胞凋亡的作用机制.方法 用不同浓度NS-398作用于人宫颈癌Hela细胞,CCK-8法检测NS-398对宫颈癌细胞增殖的影响,应用罗丹明(Rhodanmine) 123染色检测癌细胞线粒体膜电位改变;琼脂糖凝胶电泳检测DNA 降解梯度(ladder).结果 NS-398对Hela细胞的增殖具有明显抑制作用;其抑制率随药物浓度的增高、药物作用时间的延长而显著增高.NS-398作用24 h可以使线粒体膜电位显著降低且与NS-398浓度密切相关;作用48 h可以诱导Hela细胞发生凋亡.结论 NS-398对宫颈癌Hela细胞生长具有显著的抑制作用.其可能通过线粒体途径诱导癌细胞凋亡.
