小剂量阿米替林对健康志愿者血清脑-肠肽水平的影响

         

摘要

Objective To investigate the effects of low dose amitriptyline (AMT) on serum levels of brain-gut peptides. Methods This was a randomized, double-blind, placebo-controlled crossover trial, and 28 healthy volunteers completed the study. The subjects were divided into two groups, one was treated with A (AMT) 12.5 mg tid for 7 days, followed by a treatment with B(placebo), the other group was taking the oppo-site order. Fasting cubital vein blood (5 mL) were collected in the treatment(before, 7 days after and at the end). Serum levels of ghrelin, motilin, and neuropeptide Y (NPY) were measured by ELISA kits. The side effects were recorded during the treatment. Results Twenty eight healthy volunteers were successfully completed the trial. Compared with placebo, low dose AMT significantly increased the serum ghrelin level [(526.87 ± 158.44) pg/mL vs (442.87 ± 176.79)pg/mL, P=0.04] and the neuropeptide-Y level [ (965.64 ± 165.63)pg/mL vs (890.15 ± 131.46)pg/mL, P=0.03], but it had no effect on serum motilin level (P=0.61). Conclusions Low dose AMT increases the serum levels of ghrelin and neuropeptide Y, which may be related to the treatment of the functional gastrointestinal disorders.%目的:探讨小剂量阿米替林对健康志愿者血清脑-肠肽水平的影响。方法采用随机双盲交叉对照试验设计,28名健康志愿者分为2组,AB组第一周期服用A药(阿米替林),第二周期服用B药(安慰剂);BA组服药顺序相反。分别于服药前,第一周期结束,第二周期结束时抽空腹静脉血检测血清胃动素、Ghrelin及神经肽Y,并同时记录受试者的副反应。结果28名健康志愿者均完成试验。与安慰剂比较,阿米替林可显著增加血清Ghrelin、神经肽Y水平(P<0.05),但对血清胃动素水平无明显影响(P>0.05)。结论小剂量阿米替林可显著提高血清Ghrelin及神经肽Y水平,与小剂量阿米替林治疗功能性胃肠病有关。
